摘 要:目的通过对罗咪酯肽产生菌进行推理选育与抗性筛选,提高罗咪酯肽的产量。方法采用罗咪酯肽合成前体结构类似物α-氨基丁酸和快速利用氮源的结构类似物甘氨酸对罗咪酯肽产生菌进行推理诱变,同时比较以抗生素链霉素和自身抗性罗咪酯肽进行抗性诱变效果。结果在考察的4种方法中,以甘氨酸推理诱变的方法最好,正突变率高达48%,最高产量达到87.6mg/L,是原始菌株的3.5倍。自身抗性筛选的突变株最高产量65.9mg/L,是原始菌株的2.6倍。结论采用甘氨酸推理育种方法和自身抗性筛选方法,能有效提高罗咪酯肽产生菌的生产能力。Objective Rational selection and resistance screening were studied to obtain an effective breeding method of the strain, so as to improve its production of romidepsin. Methods A rational selection procedure with α-aminobutyric acid and glycine respectively as the screening agent was performed to improve the productivity of romidepsin by Chromobacterium violaceum, meanwhile the methods of streptomycin and romidepsin resistance screening were performed and the results of these four methods were compared. Result Rational selection with glycine is the most effective way among the four breeding methods. Its positive mutation rate is as high as 48%. The highest yield of romidepsin produced by the mutant strain was 3.5times higher than that of parent strain, reaching 87.6mg/L. The highest yield of romidepsin produced by the resistance screening mutant strain was 2.6 times higher than that of parent strain, reaching 65.9mg/L. Conclusion Glycine rational selection and romidepsin resistance screening can effectively improve the strain's romidepsin production.
