摘 要:Wnt/β-Catenin信号通路是广泛存在于多种真核生物中的一条高度保守的信号通路,但对于该信号通路是否参与流感病毒的复制及其对流感病毒致病性的影响目前还缺乏深入研究,为研究其对流感病毒复制的影响,本研究通过转染Wnt/β-Catenin信号通路荧光素酶报告质粒Topflash试验,表明甲型H1N1流感病毒感染后可以抑制Wnt/β-Catenin信号通路的激活。而当先转染β-Catenin蛋白真核表达重组质粒激活Wnt/β-Catenin信号通路后,再感染甲型H1N1流感病毒时,结果显示病毒复制受到了抑制,并且这种抑制作用不依赖于病毒NS1蛋白C端的PBM结构域。此外,将不同亚型流感病毒NS1蛋白真核表达重组质粒与Topflash质粒共转染,以检测其对Wnt/β-Catenin信号通路的激活能力时,结果表明H1N1的NS1可以更显著地激活Wnt/β-Catenin信号通路。本研究为进一步深入研究流感病毒感染与Wnt/β-Catenin信号通路的相互关系奠定了基础。Wnt/β-Catenin signaling pathway is highly conserved and commonly exists in eukaryotic organisms. However, the in-depth study on whether the Wnt/β-Catenin signaling pathway affects the replication and pathogenicity of influenza viruses is still lacking. In the present study, we performed a luciferase reporter transfection experiment and found that the activation of Wnt/β-Catenin pathway was inhibited by infection with H1N1 influenza virus. By contrast, we found that the replication of the H1N1 virus was inhibited when β-Catenin was transiently overexpressed before the virus infection was performed, and the inhibitory effect was not correlated with the PDZ binding motif (PBM) in the C terminus of NS1 protein. We further co-transfected the NS1 expression plasmids of different subtypes of viruses and the luciferase reporter to examine their abilities to activate the Wnt/β-Catenin pathway. We discovered that the NS1 protein of H1N1 virus is more potent in activating the Wnt/β-Catenin pathway. This study laid a foundation for further investigation of the correlation between influenza virus infection and the Wnt/β-Catenin signaling pathway.
