摘  要:利用λRed重组系统敲除鼠伤寒沙门菌SL1344的sifA基因以获取△sifA沙门菌。无菌分离培养小鼠骨髓来源巨噬细胞,分别用SL1344沙门菌及△sifA沙门菌刺激细胞,Western blot、ELISA及LDH释放检测方法检测IL-1β分泌,caspase-1表达及LDH的释放。结果表明,△sifA沙门菌促进IL-1β分泌、caspase-1表达增强、LDH释放量增多,即△sifA沙门菌增强炎性体活化。To investigate the effect of SifA in Salmonella-induced inflammasomes, in this study, we used λ Red recombination system to knockout SifA gene of Salmonella typhimurium SL1344 and obtain △sifA mutant strain. The peritoneal macrophages from mouse bone marrow wereisolated and cultured. Subsequently,cell culture supernatant and cell lysate were collected after LPS combined with different bacterial stain treated cells. Finally, ELISA, Western blot and LDH release assay were used to detect IL-1β secretion, caspase-1 expression and cell death. The results showed that △sifA promotes IL-1β secretion, caspase- 1 expression and cell death. Together, these findings demonstrated that △sifA can enhance the activation of inflammasomes.
