摘  要:畸胎瘤衍化生长因子(teratocarcinoma-derived growth factor1,TDGF1)是一种调控细胞存活、增殖、分化和迁移的小分子糖基磷脂酰肌醇(GPI)结合蛋白。TDGF1在正常组织中一般不表达或仅部分组织少量表达,在多种肿瘤细胞中表达极显著。以往有关TDGF1的报道多集中于肿瘤和早期胚胎发育,研究发现,TDGF1通过调控肌肉生成抑制素(myostatin,MSTN)信号参与成肌细胞的增殖分化。在肌源性细胞中,TDGF1的表达是CFC(Cripto-1/FRL-1/Crypic)区与ALK4(anaplastic lymphoma kinase 4)结合从而转化MSTN信号。下调TDGF1,MSTN的信号随之减弱,表明TDGF1对肌源性细胞中MSTN调控的重要性,并且EGF和CFC区对MSTN的调控都很重要。论文综述了TDGF1的分子基础研究及其对成肌细胞增殖分化的作用。TDGF1 is a small molecule sugar-based phosphatidyl inositol (GPI) binding protein for regulation of cell proliferation, survival, differentiation and migration. It is generally not in normal tissue expression but only part of the system of a small amount of expression which is expressed in a wide variety of tumor cells significantly. Previous reports of TDGF1 focused on tumor and early embryonic development. Con- versely, recent studies have found that TDGF1 through regulating the MSTN signal is involved in myo- blast proliferation and differentiation. In the cells with significant expression of Cripto, such as myogenic- cells, the CFC domain of Cripto is associated to ALK4, thereby transducing myostatin signaling. When TDGF1 is down-regulated, myostatin signaling is decreased, demonstrating the importance of TDGF1 in myostatin-controlled responses in myogenic cells. Furthermore, both the EGFand CFC domains of TDGF1 seem to be important in modulating myostatin signaling. The current review was aimed at molecular basis of the TDGF1 and its effect on differentiation of myoblast proliferation.
