摘 要:目的:探讨急、慢性发病过程中冠心病血瘀证心肌能量代谢的变化。方法:分别采用冠脉结扎法和异丙肾上腺素连续注射法结合高脂饲养+维生素D3复制大鼠冠心病急、慢性血瘀证模型,运用GC/TOF-MS对各模型组和对照组大鼠心肌组织进行代谢物检测,利用模式识别进行多元数据处理,筛选差异代谢物,并对差异代谢物进行相关通路分析,筛选差异显著的能量代谢通路。结果:冠心病急、慢性血瘀证各组大鼠心肌组织代谢谱与正常对照组明显不同,差异代谢物涉及能量代谢中重要的糖酵解、脂肪酸代谢和柠檬酸循环,代谢通路分析提示冠心病急、慢性血瘀证过程中能量代谢相关的差异显著通路有淀粉和蔗糖代谢、氨基糖和核苷酸糖代谢、柠檬酸循环、半乳糖代谢、不饱和脂肪酸代谢、氨基酸代谢、脂肪酸的生物合成等。结论:冠心病急、慢性血瘀证过程中心肌细胞能量代谢发生了变化,糖代谢、脂肪酸代谢、柠檬酸循环、氨基酸代谢均参与大鼠冠心病急、慢性血瘀证的过程。Objective: To explore the myocardial energy metabolism on acute and chronic coronary heart disease(CHD) in rats with syndrome of blood stasis based on GC/TOF-MS metabonomics. Methods: The acute and chronic CHD rat model with syndrome of blood stasis were established by high fat diet+vitamin D3 intraperitoneal injection combined with coronary artery ligation and isoproterenol injection repeatedly, respectively. The myocardial metabolites in each group were detected by GC/TOFMS, the multivariate data were analyzed by pattern recognition, and differential metabolites were identified. Consequently, key significantly changed metabolic pathways, especially energy metabolic pathways were identified according to the significantly different metabolites. Results: The myocardial metabolite profiling in group of acute and chronic CHD with syndrome of blood stasis was significantly different compared with control group. The significantly different metabolites were involved in important energy metabolic pathways, such as glycolytic pathway, fatty acid metabolism and citrate cycle. Metabolism pathway analysis indicated that several significantly different pathways were related to energy metabolism of acute and chronic CHD with blood stasis syndrome, such as starch and sucrose metabolism, amino sugars and sugar nucleotide metabolism, citrate cycle, galactose metabolism, unsaturated fatty acid metabolism, amino acid metabolism and biosynthesis of fatty acids. Conclusion: The myocardial energy metabolism including glucose metabolism, fatty acid metabolism, citrate cycle, and amino acid metabolism has changed during the process of acute and chronic CHD with syndrome of blood stasis.
