补中益气汤含药血清对TGF-β1介导的A549细胞中Vimentin及GST-π蛋白表达的影响

摘　　要：目的:观察补中益气汤含药血清对转化生长因子-β1(TGFβ1)介导的人肺腺癌A549细胞上皮间质转化过程中波形蛋白(Vimentin)及谷胱甘肽-S-转移酶π(GST-π)表达的影响。方法:制备不同浓度的补中益气汤含药血清作用于TGF-β1诱导的A549细胞,实验分组为空白血清组,空白血清+TGF-β1组以及高、中、低剂量补中益气汤含药血清+TGF-β1组。采用免疫组化、Western blot和Real-time PCR法分别检测Vimentin及GST-π的蛋白和m RNA变化。结果:高、中、低剂量的补中益气汤含药血清均能使TGF-β1诱导的上皮间质化的A549细胞中Vimentin和GST-π蛋白表达下降,m RNA表达水平降低,其中高、中剂量组与空白血清组比,差异亦有统计学意义(P<0.05,P<0.01)。结论:补中益气汤含药血清能抑制TGF-β1介导的A549细胞上皮间质化中Vimentin及GST-π的表达,改善其耐药性。Objective： To observe effects of serum containing Buzhong Yiqi Decoction on the expression of Vimentin（VIM） and glutathione S-transferaseπ（GST-π） in epithelial matter transformation process of human lung cancer A549 cells mediated by transforming growth factor-β1（TGF-β1）. Methods： The A549 cells mediated by TGF-β1 were divided into the blank serum group, blank serum＋TGF-β1 group, and high, middle, low concentration of serum containing Buzhong Yiqi Decoction＋TGF-β1 group. The changes of Vimentin and GST-π among the groups were detected by immunohistochemistry, Western blot and Real-time PCR. Results： All concentrations of serum containing Buzhong Yiqi Decoction decreased the proteins expression of Vimentin and GST in A549 cells mediated by TGF-β1, as well as the m RNA expression of vimentin and GST. Conclusion： The serum containing Buzhong Yiqi Decoction could inhibit the expression of VIM and GST-π in epithelial matter transformation process of human lung cancer A549 cells mediated by TGF-β1 and improve the its drug resistance.
