摘  要:目的:探讨神经母细胞瘤荷瘤SD幼鼠成瘤规律及其原因分析。方法:以3~4周龄SD幼鼠为研究对象,随机分组为对照组(10只)和实验组(45只根据肿瘤生长情况,又再分成瘤组和非成瘤组)。每2 d观察并且测量各组生长指标,第20天处死动物后,计算各组胸腺、脾脏脏器指数;计算外周血淋巴细胞亚群CD3^+、CD4^+、CD8^+细胞数量变化;检测各组胸腺、脾脏组织中增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)、Caspase-3蛋白的表达。结果:145只实验组SD幼鼠有13只成瘤,成瘤率为31.1%。2胸腺脏器指数:成瘤组>对照组(P<0.01),非成瘤组>对照组(P<0.01),成瘤组与非成瘤组无统计学差异(P>0.05);脾脏脏器指数在各组间无统计学差异(P>0.05)。3非成瘤组中CD3^+、CD8^+细胞数量明显高于成瘤组(P<0.01),非成瘤组与对照组、成瘤组与对照组中CD3^+、CD8^+细胞数量无统计学差异(P>0.05);CD4^+在3组中无统计学差异(P>0.05)。4胸腺组织中PCNA、Caspase-3蛋白表达:成瘤组及非成瘤组胸腺PCNA表达较对照组增加(P<0.05),Caspase-3表达较对照组降低(P<0.05),其中非成瘤组中PCNA较成瘤组明显增加(P<0.05),Caspase-3表达明显降低(P<0.05);在脾脏内PCNA、Caspase-3蛋白在各组中表达无统计学差异(P>0.05)。结论:1SD幼鼠接种人神经母细胞瘤细胞后,其生长规律出现潜伏期、快速生长期、平台期、消退期的变化;2成瘤率及成瘤规律与幼鼠免疫器官反应有关系。Objective:To explore tumorigenicity rules and cause of neuroblastoma in SD immature rats. Methods:Immature SD rats of3-4 weeks were taken as object of study and were randomly allocated into contrast group(n=10)and experimental group(n=45). After inoculating SK-N-SH cell suspension,rats were divided it into tumor group and non-tumor group. Growth indexes of all groups were observed and measured every 2 days until the animals being killed. Then viscera indexes of thymus and spleen in each group were calculated. Flow cytometry was used to calculate the number of positive cells of the peripheral blood lymphocyte subsets CD3,CD4 and CD8. Western blot was used to detect the expression of PCNA and Caspase- 3 protein of thymus and spleen tissues in all groups. Results:Among 45 SD immature rats of experimental group,13(31.1%)suffered from tumor. The weight of thymus in experimental group increased(P0.05)and there was no statistical significance between tumor group and non-tumor group(P 0.05). The weight of spleen in experimental group was the same as that in control group(P 0.05). Flow cytometry was used to examine value of peripheral blood lymphocyte subsets CD3,CD4 and CD8. CD3~+ and CD8~+ content of non-tumor group was higher than that of tumor group(P0.05). There was no difference between non-tumor group and contrast group and no difference between tumor group and contrast group(P 0.05). However,CD4 ~+ was different in the three groups(P 0.05). In thymus,expression of PCNA increased and expression of Caspase-3 decreased in experimental group(P0.05). And both the PCNA and Caspase-3 were significantly different in non-tumor group from that in tumor group(P0.05).In spleen the expression of Caspase-3 and PCNA in experimental group and control group were the same(P0.05). Conclusion:After being inoculated with human neuroblastoma cell lines,SD immature rats experiences incubation,rapid growth stage,platform stage and paracmasis. The tumor rate amd the tumorigenic
