摘  要:目的建立高效液相色谱(HPLC)法测定降脂宁有效部位中2,3,5,4’-四羟基二苯乙烯-2-O-β-D-葡萄糖苷(SBGC)和金丝桃苷的含量。方法采用大连依利特C18色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.1%甲酸溶液(15.5∶84.5);流速1.0 m L·min^(-1),检测波长340 nm,进样量20μL。结果SBGC和金丝桃苷分别在24~120μg·m L^(-1)(r=0.999 8)和2.4~12.0μg·m L^(-1)(r=0.999 6)范围内呈良好的线性关系。平均回收率分别为100.07%和100.14%,样品中SBGC和金丝桃苷的含量分别是2.26%和0.23%。结论该测定方法简便、快速、准确,可应用于降脂宁提取物中SBGC和金丝桃苷的含量测定。Objective To establish an HPLC method for determination of 2,3,5,4’-tetrahydroxystibene-2-O-β-D-glucoside(SBGC)and hyperin in the effective fraction of Jiangzhining. Methods HPLC analysis was performed on a C18 column(4.6 mm×250 mm,5 μm),with acetonitrile-0.1% methanoic acid water solution(15.5: 84.5)serving as the mobile phase. The flow rate was 1.0 mL·min^-1 ,the wavelength was 340 nm,and the injection volume was 20 μL. Results The calibration curve was linear for SBGC in the range of 24-120 μg·mL^-1(r = 0.999 8)and for hyperin in the range of 2.4-12.0 μg·mL^-1(r= 0.999 6),respectively.Their average recoveries were 100.07% and 100.14%,respectively.The contents of SBGC and hyperin were 2.26% and 0.23%,respectively Conclusion The method is convenient,precise and reliable for determination of the content of SBGC and hyperin in the effective fraction of Jiangzhining.
