摘 要:Jacalin凝集素(jacalin-related lectins,JRLs)是一类含有一个或多个JRL结构域的蛋白质,参与生物或非生物环境刺激应答.本研究基于金针菇(Flammulina velutipes)基因组数据,注释筛选到其中一个JRL基因Fv-mJRL1,对其基因结构、蛋白质性质进行了生物信息学分析,并利用其上游调控序列进一步预测到该基因潜在的转录因子Fv-Mbp1.通过实时荧光定量PCR分析得知,无论是在不同栽培基质培养的金针菇菌丝还是在金针菇各发育时期的组织中,Fv-mJRL1基因与潜在的转录因子基因Fv-Mbp1之间都存在极强的共表达规律.生物信息学及实时荧光定量PCR分析为筛选挖掘基因潜在的转录因子提供便利,后期仍需利用下游分子实验进行更为精确的验证.Jacalin-related lectins (JRLs) are a subgroup of proteins with one or more,jacalin-like lectin domains,and they participate in biological or non-biological response to environmental stimuli.The Flarnmulina velutipes genomic data were used to screen one lectin gene Fv-mJRL1 , and the gene structure and protein properties of biological information were analyzed.The potential transcrip- tion factor Fv-Mbpl was further predicted using its upstream regulatory sequence. Fluorescence quantitative RT-PCR analysis showed that Fv-mJRL1 and the transcription factor gene Fv-Mbpl were strongly co-expressed,whatever growth in different sub- stances of F. velutipes mycelium or in the various growth developmental stages. Results demonstrated that bioinformatics and qRT- PCR analysis were suitable for the screening of potential transcription factors, however,it is still necessary to use further downstream molecular experiments to verify the data.
