摘 要:目的观察中药丹参多酚酸盐对ApoE基因敲除小鼠主动脉壁血管细胞清道夫受体A和CD36mRNA表达的影响,探讨丹参多酚酸盐抗动脉粥样硬化进展的作用机制。方法 30只AopE基因敲除小鼠(4周龄,雄性)随机分成4组,即模型组(腹腔注射生理盐水,7只)、丹参多酚酸盐60 mg/kg组(7只)、丹参多酚酸盐120 mg/kg组(8只)、丹参多酚酸盐240 mg/kg组(8只),正常对照组(C57BL/6J野生型小鼠)6只。高脂饮食喂养于24周末时处死各组动物,取主动脉根部连续切片,常规苏木精-伊红染色,观察血管壁形态和脂质沉积情况,采用逆转录聚合酶链式反应对主动脉血管壁细胞清道夫受体A和CD36 mRNA表达进行定性和定量分析。结果 ApoE基因敲除鼠动脉粥样硬化斑块内均可见明显的SR-A和CD36mRNA表达,C57BL/6J小鼠正常血管内仅见少量表达;丹参多酚酸盐(240 mg/kg组)干预后,与模型组比较,ApoE基因敲除鼠动脉粥样硬化斑块内CD36 mRNA表达明显减少,差异有统计学意义(P<0.05),而SR-A mRNA的表达未见明显减少(P>0.05)。结论中药丹参多酚酸盐抗ApoE^(-/-)鼠主动脉粥样硬化可能与其下调CD36mRNA表达,减少巨噬细胞泡沫细胞的形成有关,而与SR-A无关。Objective To observe the effects of salvianolate on Apo E-knockout mice's aortic wall cell SR-A and CD36 mRNA expression,and to explore the mechanism of salvianolate's anti-atheroselerotic progress. Methods The 30 AopE knockout mice (4 weeks old,male) were randomly divided into 4 groups,namely,the model group (normal saline,Tmice), salvianolate 60 mg/kg group (7 mice), salvianolate 120 mg/kg group (8 mice), salvianolate 240 mg/kg group (8 mice),normal control group (C57BL/6J wild-type mice,6). The each group of mice were killed after 24 weeks" high-fat diet. The aortic root serial sections were taken for routine hematoxy iin- eosin staining and observing vessel wall morphology and lipid deposition;reverse transcription polymerase chain reaction was used to analyse scavenger receptor A and CD36 mRNA expression on the aorta vascular wall cell in qualitative and quantitative analysis. Results The clear expression of SR-A and CD36mRNA in ApoE knockout mice atherosclerotic plaques were visible,in C57BL/6J mice with normal blood vessels only a small amount of expression was seen;After the intervention of Salvianolate (240 mg/kg group),CD36 mRNA expression in ApoE knockout mice^s atherosclerotie plaque was significantly reduced compared with model group. The difference was statistically significant(P〈0.05),while there was no significant decrease in the SR no-A mRNA expression(P〉0.05). Conclusion The anti-atheroselerosis function of salvianolate may be associated with decreasing CD36mRNA expression and reducing macrophage foam cell formation,but not with SR-A.
