摘 要:目的探讨异氟醚对人类胚胎干细胞(hESCs)自我更新能力及分化的影响。方法将传代后1d的hESCs随机分为实验组和对照组,实验组细胞暴露于体积分数0.02异氟醚、体积分数0.05CO_2和体积分数0.21O_2的混合气体中6h,对照组细胞暴露于体积分数0.05CO_2和体积分数0.21O_2的混合气体中6h。观察hESCs的生长情况,并检测两组细胞干性标志性基因(Nanog、Sox2、Oct4、Lin28)及各胚层标志性基因(AFP、T、Nestin)的mRNA水平。结果传代后5d,实验组细胞数目明显低于对照组(t=3.40,P〈0.01);实验组细胞干性基因的mRNA水平明显低于对照组,各胚层标志性基因的mRNA水平均高于对照组(t=2.67~3.89,P〈0.05)。结论异氟醚可以抑制hESCs的自我更新能力,并促进其提前分化。Objective To explore the effects of isoflurane on self-renewal and differentiation of human embryonic stem ceils (ESCs). Methods Human ESCs--after one day of passage--were randomized to two groups as experiment group and con- trol group. The ceils in the experiment group were exposed to 2% isoflurane, 5% CO2, and 21% 02 for 6 h, and that in the con- trol group were exposed to 5% CO2 and 21% 02 for 6 h. The growth of ESCs in the two groups were observed, and the mRNA levels of sternness markers--Nanog, Sox2, Oct4, Lin28--and marker genes of three germ layers of ESCs--AFP,T and Nestin-- were detected. Results On day 5 of the passage, the number of human ESCs in the experiment groups was significantly lower than that in the control group (t = 3.40, P〈0.01). The mRNA levels of sternness markers in experiment group were lower than that in the control group and the mRNA levels of marker-genes of three germ layers were higher (t = 2.67- 3.33, P % 0.05). Conclusion Isoflurane can inhibit the self-renewal capacity of human embryonic stem cells and motivate them to he differentiated in advance.
