摘 要:目的探讨外源性磷酸肌酸对脑缺血再灌注损伤大鼠脑组织线粒体中ATP和Ca^(2+)含量影响。方法将90只Wistar大鼠随机分为假手术组、生理盐水组(对照组)和磷酸肌酸钠组(实验组)。对照组和实验组大鼠制备全脑缺血再灌注模型,并于缺血后1h分别给予腹腔内注射生理盐水和磷酸肌酸钠4mg/g,再灌注24h后断头取脑,测量脑组织线粒体中ATP和Ca^(2+)的含量。结果与对照组相比,实验组大鼠脑组织线粒体中ATP含量升高,Ca^(2+)含量降低,差异有统计学意义(F=25.54、23.07,P<0.05)。结论外源性磷酸肌酸可以改善缺血再灌注脑组织的能量代谢,并明显改善钙超载。Objective To study the effect of exogenous phosphocreatine on the content of ATP and Ca2- in cerebral mitochondria in rats following ischemia-reperfusion injury. Methods A total of 90 male Wistar rats were equally randomized to sham operation group, normal saline group(control group)and phosphocreatine group(experimental group), a model of global brain ischemia-reperfusion was created in rats in the control group and experimental group. One hour after ischemia, intraperitoneal injection of normal saline and 4 mg/g of phosphocreatine were given rats in the control and experimental group, respectively. At 24 hours after reperfusion, the brains were removed for measurement of content of ATP and Ca^2+ in mitochondrion of brain tissue. Results Compared with the control group, the ATP content in mitochondrion of brain tissue in the experimental group elevated, and Ca^2+ content declined, the difference was significant between the two groups (F=25.54,23.07;P〈0.05). Conclusion Exo- genous phosphocreatine may improve energy metabolism in ischemia-reperfusion brain tissue and calcium overload.
