固本防哮饮对信号转导及转录激活因子1及哮喘易感基因ORMDL3的影响

摘　　要：目的观察固本防哮饮对信号转导和转录因子1(STAT1)及哮喘易感基因血清类黏蛋白1样蛋白3(ORMDL3)的调控作用,分析其防治哮喘机制。方法利用白介素-4诱导人支气管上皮细胞建立哮喘细胞模型,固本防哮饮含药血清对其进行干预,采用实时荧光定量-PCR(Real Time PCR)、免疫印迹法(Western blotting,WB)、免疫荧光法(IF)分别从基因、蛋白水平检测细胞中ORMDL3和STAT1的表达及亚细胞定位。结果与正常组比较,模型组ORMDL3、STAT1 m RNA含量升高,但差异无统计学意义(P>0.05),固本防哮饮可降低模型组细胞ORMDL3、STAT1 m RNA水平,差异无统计学意义(P>0.05);与正常组比较,模型组ORMDL3、STAT1蛋白表达显著增加(P<0.05或P<0.01),与模型组相比,固本防哮饮组ORMDL3及STAT1蛋白表达均显著(P<0.05)。结论固本防哮饮对STAT1及ORMDL3的表达有一定的调控作用,可能为固本防哮饮防治哮喘的内在机制。Objective： To investigate the effect of Guben Fangxiao Decoction on STAT1 and asthma susceptibility gene ORMDL3,and analyze its mechanism. Methods： 16 HBE cells were cultured in the presence of the Th2 cytokines IL-4 for 24 hours,and then treated with Guben Fangxiao Decoction. Expressions of ORMDL3 and STAT1 were assessed by quantitative real-time PCR,Western Bloting and Immunofluorescence. Results： The expressions of ORMDL3 and STAT1 m RNA were increased after IL-4 stimulation,compared with normal 16 HBE,but the differences had no statistical significance（P 0.05）. Compared with the model group,Guben Fangxiao Decoction could decrease the expressions of ORMDL3 and STAT1 m RNA;the differences had no statistical significance（P 0.05）. Compared with the normal group,expressions of ORMDL3 and STAT1 protein of the model group were obviously increased（P 0. 05 or P 0.01）. Compared with the model group,Guben Fangxiao Decoction significantly decrease cell constitution of ORMDL3 and STAT1 protein（P 0. 05）. Conclusion： The prevention of Guben Fangxiao Decoction against asthma may be associated with regulation of STAT1 and ORMDL3.
