丹参酮ⅡA对病毒性心肌炎小鼠血清炎性因子水平的影响

摘　　要：目的观察丹参酮ⅡA对病毒性心肌炎小鼠血清肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)水平的影响。方法 80只雄性BALB/c小鼠随机分为空白对照组(对照组)20例、病毒性心肌炎模型组(模型组)30例、丹参酮ⅡA治疗组(治疗组)30例。模型组、治疗组腹腔接种柯萨奇病毒B3(CVB3)建立病毒性心肌炎模型,对照组腹腔注射不含病毒的Eagle’s液。治疗组于病毒接种0.5 h后腹腔注射丹参酮ⅡA注射液,连续14 d。观察记录小鼠一般情况、记录死亡数,计算小鼠生存率;取血测定小鼠血清IL-6、TNF-α水平;取心脏,计算心脏指数,HE染色,计算病理评分。结果与对照组小鼠比较,模型组小鼠血清IL-6和TNF-α水平、心脏指数、心脏病理评分均升高(P<0.05);与模型组比较,治疗组小鼠心脏指数、TNF-α、IL-6水平、心脏病理评分均降低(P<0.05)。结论丹参酮ⅡA可通过降低TNF-α、IL-6发挥减轻心肌损伤、治疗病毒性心肌炎作用。Objective： To study the effect of sodium tanshinone ⅡA sulfonate on tumor necrosis factor-α（TNF-α） and interleukine-6（IL-6） in mice with coxsackie B3 viral myocarditis. Methods： Eighty male BALB/c mice were randomly divided into three groups： the control group（n = 20）,viral myocarditis model group（n = 30） and sodium tanshinone Ⅱ A sulfonate-treated group（n = 30）. The first two groups were intraperitoneally inoculated with CVB3 to induce myocarditis,the control group with virus-free Eagle′s medium instead. Sodium tanshinone ⅡA sulfonate-treated group was given andrographis injection through intraperitoneal injection 0.5 hours after injecting virus every day for 14 days. The control group and myocarditis group used normal saline instead. The gen-eral state of the mice was observed;deaths were recorded and the survival rate of mice was calculated;the content of TNF-α and IL-6 was tested. Hearts were taken;the cardiac index was calculated;HE staining was adopted;the pathological integral was calculated. Results： Compared with the control group,cardiac index,TNF-α and IL-6level and the pathological integral in the model group increased（P 0.05）;compared with the model group,cardiac index,TNF-α and IL-6 level and the pathological integral in sodium tanshinone Ⅱ A sulfonate-treated group were decreased（P 0.05）. Conclusion： Sodium tanshinone ⅡA sulfonate can decrease TNF-α and IL-6serum level of mice and has obvious effects on prevention and treatment.
