通降和胃方对胃食管反流性咳嗽豚鼠呼吸道组织学形态的影响

摘　　要：目的观察通降和胃方对胃食管反流性咳嗽豚鼠模型呼吸道组织学形态的影响。方法健康雄性豚鼠40只,随机分为中药组、西药组、模型组和正常组。除正常组外,其余各组采用食管下端酸灌注法制作胃食管反流性咳嗽模型,并分别予相应药物混悬液及等量生理盐水处理,光镜下观察气管、肺病理组织学改变,透射电子显微镜下观察肺组织超微结构。结果模型组气管、肺组织炎症积分较正常组明显升高(P<0.05),中药组、西药组与模型组比较差异均有统计学意义(P<0.05);中药组气管、肺组织炎症积分低于西药组,两者比较差异有统计学意义(P<0.05)。结论通降和胃方能够有效抑制反流,降低气道神经源性炎症的发生,改善呼吸道炎症反应,达到治疗胃食管反流性咳嗽的目的。Objective： To observe the effect of Tongjiang Hewei Decoction on histopathological characteristics of the airway in guinea pigs with gastroesophageal reflux induced cough. Methods： Forty healthy male guinea pigs were randomly divided into 4 groups： Tongjiang Hewei group,the control group,the model group and the normal group. GERC models were induced repeatedly by hydrochloric acid instillation with the corresponding intervention measures to each group,except the normal group. After two weeks,all animals were executed and the histological changes of the trachea and lung tissue were observed. Evans Blue-invasive method was used to determine pulmonary capillary permeability in guinea pigs. Ultrastructural changes of lung tissues were observed under transmission electron microscope. Results： The inflammation of the trachea and lung tissue in the model group was much more obviously than that of the normal group（P 0.05）. There was a significant difference in improving the guinea pig trachea and lung tissue pathologic scores in Tongjiang Hewei group and the control group,compared with the model group. Comparing Tongjiang Hewei Decoction with omeprazole,there was a significant difference in improving the two scores（P 0.05）. Conclusions： Tongjiang Hewei Decoction can effectively inhibit acid reflux,reduce airway neurogenic inflammation,thereby reduce the inflammatory exudate and cure cough.
