摘 要:目的观察排石冲剂对高龄肾结石大鼠转化生长因子β1(TGF-β1)、肾损伤分子(KIM-1)表达的影响。方法将40只雄性高龄Wistar大鼠随机分为正常组、模型组、枸橼酸钾组和排石冲剂组,每组10只。采用1%乙二醇自由饮水和2%氯化铵灌胃建立大鼠草酸钙肾结石模型,同时各组给予相应药物,观察大鼠一般情况、体质量、24 h尿量,显微镜下观察肾组织切片中草酸钙结晶沉积,并采用酶联免疫吸附法及免疫组化法检测外周血及肾组织中TGF-β1、KIM-1的水平。结果 4周末大鼠24 h尿量比较,枸橼酸钾组和排石冲剂组均高于正常组和模型组(P<0.05);枸橼酸钾组与排石冲剂组比较,差异无统计学意义(P>0.05)。枸橼酸钾组和排石冲剂组草酸钙结晶评分均高于正常组,低于模型组(P<0.05);排石冲剂组结晶评分低于枸橼酸钾组(P<0.05)。枸橼酸钾组、排石冲剂组血清及肾组织TGF-β1、KIM-1水平明显低于模型组(P<0.05)。与枸橼酸钾组比较,排石冲剂组血清及肾组织TGF-β1、KIM-1水平显著下降(P<0.05)。结论排石冲剂能有效减少高龄肾结石大鼠TGF-β1、KIM-1的产生,减轻肾脏氧化应激及炎症损伤,抑制结石形成。Objective: To investigate the effects of Paishi Granule on TGF-β1 and KIM-1 in elderly rats with renal calcium oxalate stone. Methods: A total of 40 SPF level elderly male Wistar rats were separated into 4groups by random number table methods,including the normal group,the model group,Potassium citrate-treated group and Paishi granule-treated group(n = 10 for each). The rat model of renal calcium oxalate stone was induced by intragastrically administrated 2% ammonium chloride(AC),together with 1% ethylene glycol(EG) for free drink. At the same time,each group was treated with corresponding drugs. Individual weight and urine volume were recorded. The paraffin sections of kidney were prepared for observing the calcium oxalate deposit under the microscope. The levels of TGF-β1and KIM-1 in peripheral blood and renal tissues were detected by enzyme-linked immunosorbent assay and immunohistochemistry. Results: 24 h urine volumes in Potassium citrate-treated group and Paishi granule-treated group were significantly higher than those in normal group and model group(P 0.05),despite a non-significant difference between Potassium citrate-treated group and Paishi Granule group(P 0.05). The scores of calcium oxalate crystallization in the two treated groups were significantly higher than that in the normal group,conversely,lower than the model group(P 0.05). Furthermore,the score in Paishi granule-treated group was lower than that in Potassium citrate-treated group(P 0.05). The levels of TGF- beta 1 and KIM-1 in serum and renal tissue of Potassium citrate group and Paishi Granule group were significantly lower than those in the model group(P 0.05). Compared with those in potassium citrate group,The levels of TGF- beta1 and KIM-1 of Paishi Granule group were significantly decreased(P 0.05). Conclusion: Paishi granule can reduce the produce of TGF-β1 and KIM-1 in elderly rats with renal calcium oxalate stone,reduce renal oxidative stress and inflammatory damage,and inhibit the form
