摘  要:目的通过对中国(未包括香港、澳门特别行政区和台湾地区)脊髓灰质炎(脊灰)实验室网络(Chinese Poliomyelitis Laboratory Network,CPLN)2007年4月~2015年4月连续的细胞敏感性(Cell Sensitivity,CS)监测结果进行分析,评估其在脊灰实验室检测质量控制中的重要作用。方法全国脊灰实验室网络中所有实验室统一使用96孔微量板病毒滴定法在L20B细胞和RD细胞上对三个型别的脊灰病毒的敏感性进行检测。结果从2007~2015年,在CPLN中,有30个省级脊灰实验室已开展CS监测并按时向国家脊灰实验室上报结果,目前西藏由于实验室条件所限尚未开展CS监测。八年间的CS监测结果显示30个省级脊灰实验室的实验室质量控制(LQC)株滴度均在正常范围±0.5 logCCID_(50)/0.1ml波动。结论 2007~2015年CPLN中各个实验室所用的L20B和RD细胞对脊灰病毒敏感性处于正常范围内,细胞系状态稳定,对三个型别的脊灰病毒均敏感。CS监测作为CPLN质量控制的重要指标保证了实验室检测脊灰病毒的敏感性,为从细胞中扩增出低滴度的脊灰病毒提供了保障,对输入脊灰野病毒或疫苗衍生脊灰病毒(VDPV)的早期发现至关重要。Objective To analyze the continuous surveillance data of the cell sensitivity(CS) tests of Chinese Poliomyelitis Laboratory Network(CPLN) in China (not including Hong Kong Special Administrative Region,Macao Special Administrative Region,and Taiwan) from April 2007 to April 2015,and to evaluate its important role in the quality control of CPLN.Methods The 96-microwells plate culture and titration method were used in all polio laboratories to test the sensitivities of RD cell and L20B cell to the three serotypes of polioviruses.Results During the eight years of surveillance,30 provincial labs performed CS surveillance in CPLN(only Tibet polio laboratory did not perform CS due to the limits of laboratory conditions) and reported CS results to National Polio Laboratory(NPL) routinely.Surveillance has been continued for 8 years,and all data were ± 0.5logCCID_(50)/0.1ml of normal range.Conclusions CS to three serotypes of polioviruses for every provincial lab in CPLN was in normal range,and the cell quality was stable and sensitive.As one of the important indicators of the laboratory's quality control program,CS surveillance can ensure the ability to detect polioviruses even with a low titer;it also plays a crucial role in detecting wild polioviruses or vaccine derived polioviruses in their early stages.
