摘 要:通过调节甲叉双丙烯酰胺的浓度,制备0.1%、0.3%、0.5%、0.6%4个梯度的PAHG,并结合培养细胞用的玻璃基底,作为心肌干细胞(CSCs)粘附的基底,探索基底硬度对CSCs增殖的影响.采用组织块分离培养法,获得纯化率较高的CSCs接种到梯度PAHG上,通过EDU增殖试剂盒检测,评价基底硬度对CSCs增殖的影响.结果表明:在0.5%、0.6%的PAHG基底上,模量为15kPa左右,接近心脏生理模量18±2kPa,CSCs增殖率较高,且分化程度低.研究基底硬度对CSCs增殖影响,认识保持干细胞干性的细胞培养条件和力学环境,进而对体外分离、纯化的CSCs进行扩增,从而为满足干细胞临床移植的需求提供实验依据.0.1%,0.3%,0.6%,4,0.5% gradient PAHG were prepared by adjusting the concentration of methylene bis acrylamide,and combined with the glass substrate,as the substrate of CSCs adhesion,the effect of substrate on the proliferation of cardiac stem cells(CSCs)was explored.PAHG was inoculated into gradient CSCs with high purification efficiency by using tissue culture method.The effect of substrate hardness on the proliferation of CSCs was evaluated by EDU assay.The results showed that:on the 0.5%,0.6% of the PAHG substrate,the modulus was about 15 kPa,close to the heart physiological modulus 18 + 2kPa,CSCs proliferation rate is higher,and the degree of differentiation is low.To study the effect of substrate hardness on the proliferation of CSCs,and to recognize the cell culture conditions and mechanical environment of stem cells,and then the CSCs was amplified and purified to meet the needs of stem cell transplantation.
