摘 要:目的分析慢性热应激对大鼠肝脏mi RNA的影响。方法将Wister雄性大鼠随机分为慢性热应激组和常温对照组,提取各组大鼠肝脏组织总RNA,反转录合成c DNA,以其为模板,应用q RT-PCR方法检测大鼠肝脏组织中mir92a、mir151、mir425、mir328a、mir210、mir98的表达。结果与常温对照组相比,慢性热应激组大鼠肝脏组织中mir151、mir425、mir98、mir328a和mir210的相对表达量均明显升高(P<0.01),而mir-92a则略有升高(P>0.05)。结论慢性热应激大鼠肝脏组织中有5种mi RNA相对表达量明显升高,为日后慢性热应激标志物的寻找和抗热应激药物的研发奠定了基础。Objective To analyze the effect of chronic heat stress on mi RNA in rat liver.Methods Male Wister rats were divided into chronic heat stress and normal control groups,of which the total RNAs were extracted from liver tissues and reversely transcribed to c DNA.The expressions of mir92 a,mir151,mir425,mir328 a,mir210 and mir98 in rat liver were determined by q RT using the synthetic c DNA as a template.Results The relative expression levels of mir151,mir425,mir98,mir328 a and mir210 in chronic heat stress group increased significantly(P〈.01),while that of mir92 a increased slightly(P〈.05),as compared with those in normal control group.Conclusion The expression levels of five mi RNAs in rat liver after chronic heat stress increased significantly,which laid a foundation of finding chronic heat stress marker and developing anti-heat stress drugs.
