摘  要:目的探讨被动免疫的狂犬病抗血清和免疫球蛋白(Ig G)对小鼠的攻毒保护作用及其可能的作用机制。方法单独使用Ig G,按照125、250、1 000 IU/kg体重的剂量,分别在攻击致死剂量的强毒前后注射平均体重(20±1)g的昆明小鼠,观察14 d(脑内攻毒)或28 d(外周攻毒)后,记录小鼠发病及死亡情况,计算存活率。同时以类似的方法使用不同效价的抗血清进行试验。结果在攻毒前先注射Ig G或抗血清的条件下,小鼠的存活率与被动免疫的Ig G剂量呈正比,当Ig G剂量达1 000 IU/kg时,小鼠可得到完全保护;在脑内攻毒后注射Ig G或抗血清的条件下,无论被动免疫的Ig G剂量高低,均不能提供任何保护;但在外周攻毒3 d后注射Ig G,高剂量的Ig G可提供部分保护。结论血脑屏障可能允许部分Ig G进入脑内,并在脑内病毒感染早期阻断病毒复制,但病毒在脑内开始复制后,给予任何剂量的Ig G均不能对病毒感染提供保护。Objective To investigate the protective effect of anti-rabies sera and Ig G against virus challenge in passively immunized mice as well as the relevant mechanism.Methods Kunming mice with a mean bodyweight of(20 ± 1)g were immunized with anti-rabies Ig G at dosages of 125,250 and 1 000 IU / kg bodyweight before and after lethal virus challenge,and observed for 14(challenge by i.c.route)or 28 d(challgen by i.m.route),of which the morbidity and mortality were recorded,and the survival rates were calculated.Results The survival rate of mice immunized with Ig G or sera before virus challenge(both i.c.and i.m.)was in proportion to the dosage of Ig G / antisera,while the mice immunized with 1 000 IU / kg Ig G were completely protected.The immunization with Ig G / antisera after i.c.challenge,regardless of dosage,showed no any protective effect on mice.However,the immunization with Ig G at a high dosage 3 d after i.m.challenge provided partial protection.Conclusion The blood-brain barrier(BBB)might allow a small proportion of Ig G to enter the brain and neutralize the virus.However,once the virus in the CVS begin to replicate,the Ig G,regardless of its dose,fails to protect the mice against infection.
