摘 要:目的:探究川芎嗪(TMP)对动脉粥样硬化(AS)的作用及相关机制。方法:小鼠分为正常组、模型组、TMP组、辛伐他汀组,每组6只。高脂饲料喂养2个月后给药,同时继续给予高脂饲料至3个月,检测血清血脂(TC、TG、HDL-C、LDL-C、VLDL-C)和血管内皮生长因子(VEGF)水平,HE染色观察斑块面积,免疫荧光测主动脉CD31和vWF表达,网络药理学筛选TMP作用于血管新生的主要靶点,Western Blot检测靶点的蛋白表达。结果:与正常组比较,模型组TC、TG、LDL-C、VLDL-C升高,HDL-C降低,斑块面积增大,主动脉CD31、vWF、VEGFR2、HIF-1α、TNF-α表达升高;与模型组比较,TMP和辛伐他汀均可降低血清TG水平及主动脉CD31表达,减小斑块面积,TMP可降低VEGFR2表达,辛伐他汀可降低HIF-1α表达(P<0.05)。结论:TMP可能通过抑制VEGFR2而抑制斑块内血管新生,减小斑块面积,从而发挥抗AS的作用。Objective:To investigate the effects of Ligustrazine(TMP)on atherosclerosis(AS)and its related mechanism.Methods:Mice were divided into normal group,model group,TMP group,and simvastatin group.Mice were fed with high-fat diet for 2 months,and then continued to be fed with high-fat diet for 3 months.Serum lipids and vascular endothelial growth factor(VEGF)levels were detected.Plaque areas were observed with HE staining,expressions of CD31 and vWF in aorta were detected by immunofluorescence,main targets of TMP on angiogenesis were screened by network pharmacology.Western Blot was used to detect the expression of target protein.Results:Compared with normal group,model group had the elevated level of blood lipids,increased areas of plaque,and higher expressions of CD31,vWF,VEGFR2,HIF-1αand TNF-αin aorta.Compared with model group,TMP and simvastatin significantly decreased serum TG level and CD31 expression,and reduced plaque areas.In addition,TMP decreased VEGFR2 expression,while simvastatin decreased HIF-1αexpression(P<0.05).Conclusion:TMP might attenuate intra-plaque angiogenesis by inhibiting VEGFR2 and then reduced plaque areas,all of which plays a role in anti-arteriosclerosis.
