背/腹侧海马毁损对神经病理性疼痛大鼠的影响

摘  要:目的:观察背/腹侧海马毁损对神经病理性疼痛大鼠机械痛阈值(mechanical withdrawal threshold,MWT)的影响。方法:96只SD雄性大鼠220~250 g,行为学测定无异常后随机分成4组,A组:背/腹侧海马毁损+坐骨神经分离;B组:背/腹侧海马毁损+坐骨神经结扎(chronic constriction injury,CCI);C组:背/腹侧海马假手术组+坐骨神经分离;D组:背/腹侧海马假手术组+CCI。各组根据海马给药时间不同又分为CCI前2天给药和CCI后5天给药。每组先用立体定位手术分别给背/腹海马埋管,毁损组在特定时间给予鹅膏蕈氨酸(ibotenic acid,IBO),假手术组给予磷酸盐缓冲液(phosphate buffer solution,PBS),5天后各组分别行左侧坐骨神经结扎或分离。以各组行左侧坐骨神经结扎或分离前的机械性缩足反射阈值(MWT)为基础值,分别测量2天和7天后各组的机械痛。结果:背/腹侧海马假手术组+坐骨神经分离组,在坐骨神经分离术后第2天和第7天的MWT与基础MWT相比没有统计学差异,而背/腹侧海马假手术组+CCI组,在CCI后第2天和第7天的MWT与基础MWT相比明显下降(P〈0.05)。与腹侧海马假手术+CCI组相比,CCI前2天毁损腹侧海马在CCI后2天会引起更严重的痛觉过敏(P〈0.05),但在CCI损毁后的7天两者无统计学差异。与腹侧海马假手术+CCI组相比,在CCI后5天损毁腹侧海马,在CCI后的2天和7天两组的机械痛相近(P〉0.05)。与背侧海马假手术+CCI组相比,不管是在CCI前2天或后5天毁损背侧海马,两组在CCI后2天和7天的机械痛相近(P〉0.05)。结论:CCI能引起大鼠机械痛觉过敏,腹侧海马在神经病理性损伤的早期起抑制作用,而背侧海马在神经病理性疼痛的早期和进展中没有抑制作用。AIM: To investigate the effects of dorsal/ventral hippocampus lesion on mechanical withdrawal threshold (MWT) in neuropathic pain rats. METHODS: Ninety six adult male SD rats (220-250 g) were divided into 4 groups after past the ethological test, group A: dorsal/ventral hippo- campus lesion + sciatic nerve expose ; group B : dor- sal/ventral hippocampus lesion + sciatic nerve chro- nic constriction injury (CCI) ; group C: dorsal/ven- tral hippocampus sham operation + sciatic nerve ex- pose; group D: dorsal/ventral hippocampus sham operation + sciatic nerve CCI. Each group was di- vided into 2 sub-group according to the time point: lesion on 2d before CCI and on 5d after CCI. First, pipes were laid based on stereotaxic coordinates. Then, lesion groups were given the ibotenic acid (IBO) and sham groups were given the phosphate buffer solution (PBS) at special time. After 5d, each group exposed or ligated the left sciatic nerve after determined the MWT which were basic values. After 2d and 7d, every rats were determined the MWT. RESULTS:In group C, the MWT of 2d and 7d after exposed the sciatic nerve were similar with the basic MWT. But in group D, the MWT of 2d and 7d after CCI are significant decreased compared with the basic MWT ( P 〈 0.05 ). Compared with ventral hippocampus sham operation + CCI, ventral hippocampus lesion with IBO 2d before CCI caused mechanical hyperalgesia on 2d after CCI (P 〈 0.05), but on 7d after CCI, the MWT were no sig- nificant difference. Compared with ventral hippo- campus sham operation + CCI, the MWT of 2d and 7d after CCI were similar when ventral hippocampus lesion with IBO 5d after CCI (P 〉0.05). The dor- sal hippocampus lesion with IBO neither 2d before CCI nor 5d after CCI, the MWT of 2d and 7d after CCI were similar compared with dorsal hippocampus sham operation + CCI ( P 〉 0.05 ).
