摘 要:目的:研究西红花素对大鼠脑缺血再灌注损伤的保护作用。方法:采用大鼠脑缺血再灌注模型,隔日西红花素腹腔注射,术后每周进行神经症状评分和行为学实验。28 d后取脑检测缺血侧海马和皮层脑源性神经营养因子(BDNF)、胶质细胞源性神经营养因子(GDNF)、丙二醛(MDA)含量和超氧化物歧化酶(SOD)、谷胱甘肽过氧化酶(GSH-Px)活性。结果:西红花素给药显著降低脑缺血再灌注大鼠的神经症状评分,增加十字高架迷宫试验的开臂停留时间,减少旷场试验中央区活动时间。同时,西红花素腹腔注射明显上调缺血侧海马和皮层GDNF、皮层BDNF的表达,降低MDA的产生,提高SOD、GSH-Px活性(P<0.05),差异具有统计学意义。结论:西红花素可缓解脑缺血再灌注大鼠的焦虑情绪,提高空间辨识和学习记忆能力,其机制可能与西红花素促进神经营养因子的分泌和抗氧化活性物质的产生有关。AIM: To observe the protective effect of crocin on cerebral ischemia-reperfusion in- jury in rats. METHODS: Crocin (25, 50, 100 mg/kg) was i.p. injected every 2 days after focal cerebral ischemia for 90 min and reperfusion in rats. The neurological deficits and neurobehavioral tests were determined once a week. The content of BD- NF, GDNF, MDA and the activity of SOD, GSH-Px in hippocampus and cortex of the ischemia side was detected 28 d after surgery. RESULTS:Crocin sig- nificantly reduced the nerurological deficits, in- creased residence time in open arm of elevated plus maze test and in the middle area of open field test (P 〈0. 05). Crocin markedly up regulated the pro- duction of BDNF, GDNF, MDA and the activity of SOD, GSH-Px in hippocampus and/or cortex of the ischemia side. CONCLUSION: Crocin could re- lease the anxiety of rats after cerebral ischemia- reperfusion injury and improve spatial learning and memory, which may relate to the increased secretion of neurotrophic factors and antioxidant property of crocin.
