当归芍药散含药血清对ET-1诱导的HSC-T6细胞收缩的影响

摘　　要：目的:通过体外实验研究当归芍药散含药血清对ET-1诱导的肝星状细胞(HSC-T6细胞)收缩的作用。方法:采用FITC-鬼笔环肽细胞骨架染色方法观察HSC-T6细胞肌动蛋白丝变化的数量;采用I型鼠尾胶原法观察HSC-T6细胞收缩的影响。结果:当归芍药散含药血清各剂量组能有效抑制肌动蛋白丝数量增多;在第24 h,模型组(ET-1)的胶原凝胶面积明显缩小(P<0.01);当归芍药散含药血清各剂量组可以不同程度地抑制ET-1(10 nmol/L)引起的HSC-T6细胞胶原凝胶收缩(P<0.05)。结论:当归芍药散含药血清可以抑制ET-1诱导的HSCs收缩。AIM： To research the effects of medicated serum of Dangguishaoyao San on HSC-T6 cells contraction induced by endothelin-1 in vitro.METHODS： The quantity changes of actin filament of HSC-T6 cells were observed with FITC- phalloidin staining; the cell contraction effects of HSC-T6 cells were evaluated using Rat tail collagen type I method. RESULTS： The actin filament quantity was effectively inhibited in each dose group of medicated serum of Dangguishaoyao san and Y-27632; area of collagen gel was calculated by image analysis system. After 24 h,compared with the control group,the collagen gel area of model group（ ET-1） was significantly shrunk（ P〈 0. 01）; HSC-T6 cell contraction induced by ET-1（ 10 nmol / L） was shrunk in different extent by medicated serum of Dangguishaoyao san in each dose group of（ P〈 0. 05）; Y-27632 inhibition group also possessed the similar effect（ P 〈0. 01）. CONCLUSION： Medicated serum of Dangguishaoyao San could inhibit the HSCs contraction induced by ET-1,thus reducing intrahepatic vascular resistance and portal pressure.
