Hsp70对慢性心理应激小鼠胚胎在体外培养中的作用

摘　　要：目的探讨HSP70能否对慢性轻度不可预见性应激(CUMS)小鼠2-细胞胚胎在体外培养的环境中起到应激保护作用。方法设立对照组,构建CUMS雌性昆明小鼠模型,予以PMSG/HCG促排卵获取2-细胞胚胎,置于M16培养液中培养48小时,观察优胚率及囊胚形成率,免疫荧光检测Hsp70及Caspase-3在胚胎的表达情况,共培养囊胚与Ishikawa细胞观察粘附率,并进行统计学分析。结果实验组的优胚率和囊胚率分别为62.2%和20.6%,均低于对照组的89%和31%,差异有统计学意义(P<0.05)。HSP70在实验组胚胎中阳性表达率为86.67%,高于对照组的30%,差异有统计学意义(P<0.05)。Caspase-3在实验组及对照组之间阴性表达率无统计学差异(P<0.05)。实验组胚胎的粘附率为20%,低于对照组的30%,差异有统计学意义(P<0.05)。结论 HSP70可能对CUMS小鼠2-细胞胚胎在体外培养的环境中起到应激保护作用,该作用可能通过抗凋亡因子(caspase-3)实现。Objective To explore whether HSP70 can play aprotective role in 2-cells in vitro in the mouse with chronic unpredictable mild stress（CUMS）.Methods We established an animal model of CUMS using female Kunming mice and administrated PMSG/hCG for induction of superovulation.After obtained 2-cell embryos,we cultured them in M16 culture medium for 48 hours and observed the rate of good quality embryos and blastocyst formation.We detected the expression of HSP70 and Caspase-3in the embryos by immunofluorescence method and calculated the adhesion rate of embryos to endometrial epithelial Ishikawa cells in vitro.Results The good quality embryo rate and blastocyst formatiaon rate of the experimental group were 62.2% and 20.6%respectively.Both of which were significantly lower than that of the control group（89% and 31%,P〈0.05）.The positive expression rate of HSP70 in the experimental group was86.67%,which was significantly higher than that in the control group（30%,P〈0.05）.There was no significant difference in both groups about the negative expression rate of Caspase-3.When cultured the embryos with Ishikawa cells for 48 hours,we observed that the adhesion rate of the experimental group was 20%,which was lower than that of the control group（30%,P〈0.05）.Conclusion HSP70 may play aprotective role in 2-cells of CUMS mouse in vitro,which may be realized by anti apoptotic factors（caspase-3）.
