外源性高迁移率族蛋白B1预处理对大鼠心肌缺血再灌注损伤的保护作用

摘  要:目的探讨外源性高迁移率族蛋白B1(HMGB1)预处理对大鼠心肌缺血再灌注损伤的影响。方法选择健康雄性SD大鼠60只,随机分为5组,假手术组、缺血再灌注组、预处理组、LY294002组、SB203580组,每组12只。建立大鼠心肌缺血再灌注模型。检测各组大鼠心肌梗死范围、肌酸激酶、乳酸脱氢酶、TNF-α、白细胞介素6、超氧化物歧化酶、丙二醛的表达变化。结果与缺血再灌注组比较,预处理组心肌梗死面积显著缩小(P<0.05);与预处理组比较,LY294002组和SB203580组大鼠心肌梗死面积明显增大[(47.5±3.0)%,(44.6±2.8)%vs(32.2±2.5)%,P<0.05]。与假手术组比较,缺血再灌注组乳酸脱氢酶、肌酸激酶、白细胞介素6、TNF-α及丙二醛水平明显升高,超氧化物歧化酶水平明显降低(P<0.05);与缺血再灌注组比较,预处理组乳酸脱氢酶、肌酸激酶、白细胞介素6、TNF-α及丙二醛水平明显降低,超氧化物歧化酶水平明显升高(P<0.05);与预处理组比较,LY294002组和SB203580组乳酸脱氢酶、肌酸激酶、白细胞介素6、TNF-α及丙二醛水平明显升高,超氧化物歧化酶水平明显降低(P<0.05)。结论外源性HMGB1预处理对大鼠心肌缺血再灌注损伤具有显著保护作用。Objective To study the protective effect of exogenous HMGB1 protein preconditioning on rat myocardial I/R injury and its mechanism.Methods Sixty healthy male SD rats were divided into sham operation group,I/R group,HMGB1 protein pretreatment group,LY294002 group and SB203580group(12in each group).A rat myocardial I/R model was established.The myocardial infarction size was measured and expressions of CK,LDH,TNF-α,IL-6,SOD and MDA were detected.Results The myocardial infarction size was significantly smaller in HMGB1 protein pretreatment group than in I/R group(P〈0.05)and was significantly larger in LY294002 group and SB203580 group than in HMGB1 protein pretreatment group(P〈0.05).The expression levels of CK,LDH,TNF-α,IL-6and MDA were significantly higher whereas the expression level of SOD was significantly lower in I/R group than in sham operation group(P〈0.05).The expression levels of CK,LDH,TNF-α,IL-6and MDA were significantly lower whereas the expression level of SOD was significantly higher in HMGB1 protein pretreatment group than in I/R group(P〈0.05).The expression levels of CK,LDH,TNF-α,IL-6and MDA were significantly higher whereas the expression level of SOD was significantly lower in LY294002 group and SB203580 group than in HMGB1 protein pretreatment group(P〈0.05).Conclusion Exogenous HMGB1 protein preconditioning can effectively protect rats against myocardial I/R injury.
