摘 要:为探讨免疫增强剂(VA5)提高SPF鸡免疫功能的机理,将SPF鸡分别免疫H9亚型禽流感灭活疫苗、含VA5的H9灭活疫苗、含IL-4和/或IFN-γ真核质粒的H9亚型禽流感灭活疫苗,另设空白对照。检测免疫鸡外周血CD4^+、CD8^+T淋巴细胞亚群数目及IL-4和IFN-γ细胞因子含量的变化及血清抗体效价。结果显示,在14日龄、17日龄和21日龄和28日龄,免疫IL-4和H9、IL-4与IFN-γ和H9以及VA5佐剂与H9组的CD4^+淋巴细胞亚群上升快且比例高于其他组,免疫IFN-γ和H9、IL-4与IFN-γ和H9以及VA5增强剂与H9组的CD8^+淋巴细胞亚群上升快且比例高于其他组,IL-4的上升趋势与CD4^+细胞类似,IFN-γ的上升趋势与CD8^+类似。免疫含VA5的禽流感(H9亚型)灭活苗组在免疫后第2周、3周和4周血清HI效价达到7lg2以上,高于其他各免疫组。说明,与直接添加IL-4和IFN-γ的细胞因子质粒组相比,VA5免疫增强剂能够促进SPF鸡外周血淋巴细胞增值,提高CD4^+、CD8^+T淋巴细胞的比例,并诱导IL-4和IFN-γ细胞因子分泌。The aim of this study is to explore the immunological mechanism of immunopotentiator VA5 in improving the immune response of specific pathogen-free ( SPF ) chickens. SPF chickens were challenged with the inactivated H9 subtype vaccine, VA5+H9 vaccine, IL?4+H9 vaccine, IFN?γ+ H9 vaccine, IL?4 + IFN?γ + H9 vaccine, and blank control, respectively. The number of CD4+ or CD8+subgroup lymphocytes from peripheral blood mononuclear cells, the contents of IL?4 and IFN?γ from sera and the antibody levels of hemagglutinin inhibition ( HI) were detected. The number of CD4+ subgroup lym? phocytes from chickens in groups of IL?4+H9, IL?4+IFN?γ+H9, and VA5+H9 climbed faster and the percentage was higher than those in other groups at the 14?, 17?, 21? and 28?day ages. The number of CD8+ subgroup lymphocytes from chickens in groups of IFN?γ+H9, IL?4 and IFN?γ+H9 vaccine and VA5+H9 vaccine climbedfaster and the percentage was higher than those in other groups IL4 showed similar rising trend to the CD4+ subgroup lymphocytes and IFN?γ presented similar rising trend to the CD8+subgroup lymphocytes. The HI antibody levels of chickens in group of VA5+H9 were 7lg2 at 2?, 3?and 4?week postvaccination, higher than those in other groups. In summary, the immunopotentiator VA5 increased the percantages of CD4+ and CD8+ subgroup lymphocytes and induced high secretions of IL?4 and IFN?γ.
