复方五指毛桃稠膏的制备工艺、质量标准及抗氧化活性研究

摘　　要：目的:五指毛桃单味药材及其复方提取工艺研究;复方质量标准、抗氧化活性研究。方法:单味药材分别采用95%乙醇提取(A)、水提取(B)、水中加入猪肉提取(C);补骨脂素的含量采用高效液相色谱法(High-performance liquid chromatography,HPLC)测定;多糖含量采用紫外分光光度法测定;抗氧化活性采用氧化时间法、总还原力进行综合评价。结果:A、B、C三种提取方法,分别可提取出0.469、0.098、0.387 mg补骨脂素;每100 g稠膏的多糖含量分别为0、5.29、3.12 g。故确定复方的提取工艺:五指毛桃先醇提,再与复方合并水提。补骨脂素、水溶性多糖含量分别为0.383 mg/g药材、31.5 g/100 g稠膏。复方稠膏质量标准中:五指毛桃、茯苓、大枣采用薄层色谱(Thin-layer chromatography,TLC)进行定性分析;HPLC对补骨脂素进行定量分析。抗氧化能力结果显示:复方>C>B>A。结论:采用先醇提后水提的制备工艺,能有效提出五指毛桃中的多种有效成分;与单味药材相比,组成复方后抗氧化能力增强,可为进一步的食品开发提供依据。Purpose： To investigate the extraction preparation,quality standard and antioxidant activity of Ficus hirta Vahl,with or without other herbs.Methods： Ficus hirta Vahl was extracted by three different solutions,which there were alcohol,water and broth respectively,corresponding mathod A,B and C.The psoralen and polysaccharide content of different methods were compared.Psoralen was analyzed by high-performance liquid chromatography（HPLC）.Polysaccharide was evaluated by UV spectrophotometer.The oxidation time and reducing power were used to evaluate antioxidant activities comprehensively.Results： Psoralen in per gram of Ficus hirta Vahl were 0.469 mg（A）,0.098 mg（B） and 0.387 mg（C）.Polysaccharide content in per hundred gram of extraction were 0g（A）,5.29 g（B） and 3.12 g（C）.Then the extraction preparation of Ficus hirta Vahl with other herbs was as follows： Ficus hirta Vahl was extracted by 95% alcohol and the solution were filtrated.The filter risidue with other herbs were extracted by water.And the extraction of psoralen was 0.3825 mg/g and polysaccharide was 31.5 g/100 g.The qualitative analysis includes Ficus hirta Vahl,Poria cocos,Fructus Ziziphi Jujubae was by thin-layer chromatography（TLC）.And the quantitative analysis of psoralen was by HPLC.Antioxidant activity： Ficus hirta Vahl with other herbsCBA.Conclusion： It is an effective way to extract different polar active ingredients from Ficus hirta Vahl by alcohol or water.Ficus hirta Vahl with other herbs have more antioxidant activity than itselve＇ s only.
