摘  要:目的 探讨优化处理后精子的最佳储存条件.方法 收集禁欲2-7 d的精液标本124例,所有样本均采用密度梯度离心法进行处理,将处理后的精子分别放置室温(20℃左右,18-25℃)、35℃恒温水浴箱和37℃6%CO2培养箱,放置4h和24 h后采用计算机辅助精液分析系统(CASA)检测精子前向运动百分率以及各级精子运动百分率.结果 35℃恒温箱组放置4h后精子前向运动率和a级精子运动百分率高于室温组和37℃6%CO2培养箱组[精子前向运动率:(75.34±6.87)%比(66.69±16.11)%、(71.46±11.89)%,a级精子运动百分率:(44.80±12.76)%比(31.08±11.51)%、(34.42±9.90)%,均P<0.05],而在室温组和37℃6%CO2培养箱组比较差异则无统计学意义(均P>0.05).放置4hc级精子百分率室温组要高于35℃恒温箱组和37℃6%CO2培养箱组[(14.02±6.44)%比(7.33±1.82)%、(8.58±4.52)%,均P<0.05)],而在35℃恒温箱组和37℃6%CO2培养箱组比较差异则无统计学意义(均P>0.05).放置4h3组间b级和d级精子百分率比较差异无统计学意义(均P>0.05).处理后精子放置24h后各组间前向运动精子百分率和各级精子百分率差异均无统计学意义(均P>0.05).结论 精液处理后放置35℃恒温箱中的精子运动参数相对较好.Objective To investigate the optimal storage conditions for processed sperms.Methods A total of 124 semen samples were collected after 2 to 7 d of abstinence.All samples were processed with density gradient centrifugation.After this,the processed sperms were placed in room temperature (around 20 ℃,18-25 ℃),35 ℃ water bath and 37 ℃ 6% CO2 incubation,respectively.At 4 h and 24 h later,computer-assisted semen analysis (CASA) system was used to determine the percentage of sperms with forward movement as well as the percentage of sperm motility according to different grades.Results At 4 h,sperms placed in 35 ℃ incubator showed significantly higher percentages of forward movement and grade A sperm motility compared with sperms in room temperature and 37 ℃ 6% CO2 incubator [forward movement:(75.34t6.87)% vs (66.69± 16.11)%,(71.46± 11.89)%;Grade a sperm motility:(44.80±12.76)% vs (31.08±11.51)%,(34.42±9.90)%,all P〈0.05].These data did not differ significantly between the room temperature group and the 37 ℃ 6% CO2 incubator group (all P〉0.05).At 4 h,the percentage of Grade C sperm motility in room temperature group was higher than that in the 35 ℃ incubator group or the 37 ℃ 6% CO2 incubator group [(14.02±6.44)% vs (7.33±1.82)%,(8.58±4.52)%,all P〈0.05],but did not differ significantly between the 35 ℃ incubator group and the 37 ℃ 6% CO2 incubator group (all P〉0.05).At 4 h,the three groups did not show statistically significant differences in Grade B or D sperm motility (all P〉0.05).At 24 h,all groups of sperms did not differ in percentage of forward movement and sperm motility by any grades (all P〉0.05).Conclusion Processed semen placed in 35 ℃ incubator shows comparatively better performance of sperm motility.
