摘 要:目的建立库仑阵列电化学-高效液相色谱法同时测定大鼠海马、皮质、下丘脑中8种单胺类神经递质的分析方法。方法采用库仑阵列电化学高效液相色谱仪检测,以内标法测定大鼠不同脑区中单胺类神经递质及其相关代谢产物的含量。结果 8种单胺类神经递质在本实验条件下线性关系良好,最低检测限在0.245~0.330μg/L之间,平均回收率均在87.0%~98.3%之间,大鼠不同脑区中8种单胺类神经递质的含量均在28.2~2472.8 ng/g之间。结论该方法具有操作简便、快速、准确、灵敏、分离度好、仪器稳定等优点,可以用于大鼠不同脑区8种单胺类神经递质的同时检测与分析。Objective To establish a method for simultaneous detection of monoamine neurotransmitters and their metabolites in the different sections of rat brains with HPLC and coularray electrochemistry. Methods HPLC-ECD was used for detection,and the monoamine neurotransmitters were quantitatively analyzed by internal standard method. Results Under the optimized separation conditions,the calibration curves showed good linearity. The lowest detectable limit was between 0.245-0.330 μg/L,the average recovery rate was between 87.0%- 98.3% and the content of monoamine neurotransmitters in different brain regions of rats was between 28.2-2472.8 ng/g. Conclusion The method has the advantages including easy and prompt operation,accuracy,sensitiveness,good resolution and stabilized instrument,and can be used for simultaneous detection and analysis on 8 monoamine neurotransmitters in rat brains.
