摘 要:本研究以脂肪沉积能力超强的阿勒泰羊为研究对象,探究绵羊PLA2G16基因多态性并分析其对氨基酸序列和蛋白高级结构的影响,为后续深入研究该基因在绵羊脂肪代谢中的作用机制奠定基础。采用PCR—SSCP技术对阿勒泰羊PLA2G16基因第4外显子的多态性进行了检测,并分析了氨基酸序列和高级结构的变化特征。结果表明,阿勒泰羊PLA2G16基因第4外显子处存在一个G〉A突变,该突变使原来的密码子GAG变为AAG,从而使PLA2G16蛋白第111位的氨基酸残基由谷氨酸变为赖氨酸.蛋白质高级结构预测软件显示该突变可以引起蛋白结构的变化,这可能影响其正常的生物学功能。该结果为进一步探讨PLA2G16基因在阿勒泰羊脂肪代谢中的作用机制,培养低脂肪绵羊品种积淀了数据基础。To investigate the polymorphism of sheep PLA2G16 gene and its effect on the sequences and structure of PLA2G16 protein, and to lay the foundation for further understand its function in sheep fat deposition. Here Ahay sheep with super strong fat- storing ability were chosen as the research subject, and PCR-SSCP method was employed to detect the polymorphism of the fourth exon of PLA2G16 gene, and then the change of amino acid sequence and protein structure were further analyzed. The result showed that there was one G〉A mutation in the fourth exon of PLA2G16 gene, and this mutation caused a missense mutation of Glu-~Lys in the 11 lth amino acid of PLA2G16 protein. The structure of PLA2G]6 also changed according to the calculation of Phyre2 on line soft ware and its bio-function maybe alter correspondingly. These results may help to explore the role of PLA2G16 gene in Altay sheep fat deposition, and then to accumulate fundamental data for low-fat sheep breeding.
