摘  要:茉莉酸(JA)在植物生殖发育中发挥重要调控作用,尤其是促进禾本科植物的颖花开放。然而迄今有关高粱颖花开放削浆片内JA信号相关基因的表达变化模式与颖花开放时间的关系尚不清楚。以常规高粱种质"625R"为试材,采用RNA-seq技术分析高粱颖花开放前12 h和1 h的浆片转录组差异;并通过与数据库比对,分析高粱颖花开放前茉莉酸生物合成及信号转导途径相关基因的表达变化。结果表明,2个时期浆片组织中富集于茉莉酸生物合成及信号转导途径的差异基因有51个,且以花前12 h为对照,上调基因有41个,下调基因10个。茉莉酸生物合成途径中,差异基因主要涉及JA合成的上游调控步骤,而JA信号转导途径中则以JAZ蛋白响应基因数目最多、强度最大。研究结果为进一步阐明颖花开放进程中浆片吸水膨大的分子机制提供依据。Jasmonic acid( JA) is an important phytohormone in regulating plant reproductive development,especially in the inductive effect on the floret opening in grasses. However,The relationship between the time of floret opening and the expression changes patterns of those genes involved in JA signaling in lodicules of sorghum remained unclear.In this study,the transcriptomes differences of lodicules from 12 h and 1 h before floret opening were analyzed using RNA sequencing technology,in which the experimental materials came from the Sorghum germplasm( Sorghum bicolor.cv.625R).The changes of expression of JA biosynthesis and JA signal transducer pathways in Lodicules before floret opening in Sorghum were studied by comparing them with databases.The findings of this study showed that 51 differentially expressed genes identified from transcriptome data were referred to jasmonic acid( JA) biosynthesis and JA signal transducer pathways,and comparing them with Sor-12 h,41 DEGs of which were up-regulated with the approach of the flowering.Moreover,the differentially expressed genes were involved in the protein of upstream step of the pathway of JA biosynthesis. And the number of genes coding for JAZ was the most and the strength of them was the biggest in JA signal transduction pathway.The results laid the foundation for further study of the molecular mechanism of lodicule cell expansion in the process of floret opening in sorghum.
