摘 要:目的:测定金线吊乌龟多糖的含量。方法:采用水提醇沉法提取、纯化金线吊乌龟多糖,以葡萄糖标准液绘制标准曲线,用硫酸-苯酚比色法于490 nm波长处测定金线吊乌龟多糖的含量。结果:金线吊乌龟精制多糖得率为2.31%,精制多糖含量为71.78%,金线吊乌龟原药材中总多糖含量为1.66%。结论:利用硫酸-苯酚比色法测定金线吊乌龟多糖含量,简便易行,稳定性、精密度和重复性良好,结果准确。Objective: To detect the content of polysacch.afide from Jinxiandiaowugui. Method: The polysaccharide from Jinxiandiaowugui was extracted and purified by water-extraction and alcohol-precipitation method. The standard curve was established using glucose standard solution. Sulfuric acid-phenol colorimetric method was applied to detect polysaccharide content from Jinxiandiaowugui at the wave length of 490nm. Result: The yield rate of refined polysaccharide from Jinxiandiaowugui was 2.31%. The content of refined polysaccharide was 71.78%. The content of total polysaccharide from original medicinal materials of Jinxiandiaowugui was 1.66%. Conclusion: The sulfuric acid-phenol eolorimetric method used in detection of polysaecharide content from Jinxiandiaowugui is simple and rapid. The result is accurate with high stability, precision and repeatability.
