摘  要:目的:建立HPLC-DAD同时测定白花蛇舌草中齐墩果酸和熊果酸含量的方法。方法:采用Spursil C18色谱柱(4.6×250 mm,5μm),流动相为甲醇-水-冰醋酸-三乙胺(90∶10∶0.03∶0.06),流速0.5 m L﹒min^-1,柱温35℃,检测波长210 nm,进样量10μL。结果:齐墩果酸和熊果酸分别在0.0218-0.218μg﹒μL^-1(r=0.9997)和0.07312-0.7312μg﹒μL^-1(r=0.9996)范围内线性关系良好,平均回收率分别98.17%(RSD=1.89%)、99.31%(RSD=2.62%)。结论:HPLC-DAD能同时测定白花蛇舌草中齐墩果酸和熊果酸含量,并且该检测方法简便准确、稳定可靠,可用于白花蛇舌草药材的质量控制。Objective: To establish a HPLC-DAD method for simultaneous determination of oleanolic acid and ursolic acid in Baihuasheshecao. Method: The separation was performed on the Spursil C18 column (4.6 mm×250 mm, 5 μm) with methanol- water-glacial acetic acid-triethylamine (90:10:0.03:0.06) as mobile phase. The detection wavelength was 210 nm. The flow rate was 0.5 mL· min^-1. The column temperature was 35 ℃, and the injection volume was 10 μL. Result: The linear response of oleanolic acid and ursolic acid were 0.0218-0.218μg·μL^-1 (r=0.9997) and 0.07312-0.7312 μg· μL^-1 (r=0.9996) respectively. The average recovery of oleanolic acid and ursolic acid were 98.17% and 99.31%, with RSD of 1.89% and 2.62% respectively. Conclusion: HPLC-DAD can be used to simultaneously detect oleanolic acid and ursolic acid in Baihuasheshecao. The detection method is simple, accurate, reliable and can be used for quality control of Baihuasheshecao.
