黄连素对IEC-6细胞株增殖与凋亡的影响

摘  要:目的观察黄连素对IEC-6细胞增殖和凋亡的影响。方法采用水溶性四氮唑^(-1)(WST^(-1))比色法观察黄连素对IEC-6细胞增殖的影响;采用Annexin V-FITC/PI双标流式细胞术检测黄连素对IEC-6细胞凋亡的影响。结果与正常胎牛血清培养组比较,黄连素低浓度组细胞存活率显著增加,其中1μg·m L^(-1)黄连素组促增殖作用最显著,细胞存活率为220.17%,故选用1μg·m L^(-1)黄连素浓度作为工作浓度。流式细胞术检测结果显示,黄连素干预24 h后,不同葡萄糖状态下细胞的凋亡率均明显下降。与高糖组比较,黄连素高糖组细胞凋亡率下降了6.23%;与低糖组比较,黄连素低糖组细胞凋亡率下降了8.51%;与超高糖组比较,黄连素超高糖组细胞凋亡率下降了9.30%。结论黄连素能够促进肠道L细胞系IEC-6细胞株增殖,并减少其凋亡。Objective To observe the effects of berberine on the proliferation and apoptosis of IEC- 6 cells. Methods The proliferation of IEC- 6 cells was observed by water- soluble tetrazolium^(-1)(WST^(-1)) assay. The apoptosis of IEC- 6cells was observed by flow cytometry through fluorescein annexin V- FITC/PI double labeling. Results The viability of IEC- 6 cells was significantly increased after intervention with berberine as compared with normal culture group.Berberine at 1 μg·m L^(-1)had the strongest effect on promoting the proliferation of IEC- 6 cells,with a survival rate of220.17 %. Therefore, the working concentration of berberine solution was named as 1 μg·m L^(-1). The flow cytometry results showed that apoptotic rate of IEC- 6 cells at different concentrations of glucose decreased significantly after berberine intervention for 24 h. Compared with high glucose group,the apoptotic rate of high glucose+berberine group decreased by 6.23 %. Compared with the low glucose group, the apoptotic rate of low glucose+berberine group decreased by 8.51 %. Compared with the super- high glucose group, the apoptotic rate of super- high glucose+berberine group decreased by 9.30 %. Conclusion Berberine could promote the proliferation of IEC- 6 cells, and reduce the apoptosis of IEC- 6 cells.
