摘  要:目的考察新疆两色金鸡菊乙酸乙酯(EAE)和正丁醇(BE)提取物对自由基清除能力和醛糖还原酶的抑制活性。方法以叔丁基对羟基茴香醚(BHA)和维生素C(Vitamin C)为阳性对照,分别测定提取物对DPPH、ABTS、羟自由基(*OH)和超氧阴离子(*O_2^-)等自由基的清除能力;以槲皮素为阳性对照,DL-甘油醛为底物,醛糖还原酶(来源于大鼠眼晶状体)和辅酶NADPH组成的反应体系,测定提取物抑制醛糖还原酶的活性。结果与阳性对照BHA和Vitamin C比较,两色金鸡菊提取物对DPPH、ABTS、*OH和*O_2^-等自由基有不同程度的清除作用;与阳性对照药物槲皮素相比,提取物对醛糖还原酶的抑制能力用IC_(50)值表示,差异有统计学意义(P<0.05),提取物对醛糖还原酶有一定的抑制作用但弱于槲皮素。结论新疆两色金鸡菊乙酸乙酯和正丁醇提取物有清除自由基的能力,同时具有一定抑制醛糖还原酶的活性。Objective To investigate the free radical- scavenging capacity and aldose reductase inhibitory activity of ethyl acetate extract(EAE) and n- butanol extract(BE) from Coreopsis tinctoria Nutt. in Xinjiang. Methods With butylated hydroxyanisole(BHA) and Vitamin C as the positive controls, we investigated the free radical- scavenging properties of the EAE and BE on DPPH, ABTS, hydroxyl free radical(*OH) and superoxide anion radical(*O_2^-).With quercetin as the positive control, the aldose reductase inhibitory activity was assayed by using reaction system including DL- glyceraldehyde(using as substrate), the aldose reductase(from rat lens) and NADPH(for coenzyme).Results Compared to the BHA and Vitamin C, EAE and BE had certain radical- scavenging capacity for scavenging DPPH,ABTS,*OH,and *O_2^-. The aldose reductase inhibitory activities(IC_(50))of EAE and BE were different from those of the quercetin(P〈0.05). The EAE and BE had certain aldose reductase inhibitory effect,but their activities were weaker than the quercetin. Conclusion The EAE and BE of Coreopsis tinctoria Nutt. have certain free radical- scavenging capacity and aldose reductase inhibitory activity.
