摘 要:目的观察别旁茶(BPC)总苷对溃疡性结肠炎(UC)小鼠模型干预的药效作用,考察不同分组小鼠的尿液代谢表达谱的差异。方法 balb/c小鼠随机分为BPC组、三硝基苯磺酸(TNBS)干预组、正常对照组和阳性药组,药物干预后测定药效指标,采用超高效液相色谱-四级杆飞行时间质谱(UPLC/Q-TOf MS)分析动物尿样,数据处理采用主成分分析(PCA)和偏最小二乘判别法(PLSDA)。结果与模型组比较,BPC组小鼠结肠组织损伤明显减轻,SOD活性升高(P<0.01),MPO活性和MDA水平明显降低(P<0.01)。代谢组学模式识别分析各组区别明显,PLSDA方法区分尤为明显。结论代谢组学初步研究显示,别旁茶总苷可明显改善溃疡性结肠炎小鼠模型的病变,PLSDA代谢组学数据处理方法可靠。Objective to study the intervention effect of total glycosides from Jasminum elongatum(P.J.Bergius) Willd.(JEW- TP),a herbal medicine used by Yao nationality,on mice model of ulcerative colitis(UC),and to investigate the differential expression profiles of urine metabolism from different groups of mice. Methods Balb/c mice were randomly divided into JEW- TP group, model group, normal control group, and positive control group. The biochemical indicators of mice urine were measured with ultra- high performance liquid chromatography/quadrupoletime- of- flight mass spectrometry(UPLC- Q- TOF/MS) after administration of the drug. And then the biochemical data were processed with principal components analysis(PCA) and partial least squares discriminant analysis(PLSDA).Results Compared with the model group, JEW- TP group significantly reduced mice colonic injury, increased SOD activity significantly(P〈0.01), and lowered MPO activity and MDA level significantly(P〈0.01). Application of metabolomics mode could be easy to identify the differences among the groups, and the distinguishing results were better when used with PLSDS. Conclusion Preliminary metabolomics study shows that total glycosides from JEW can significantly relieve the colonic injury of ulcerative colitis mice, and PLSDA for the metabolomics data processing is reliable.
