摘  要:目的建立岗梅种子的检验方法。方法参考《国际种子检验规程》和《农作物种子检验规程》的方法,通过对岗梅种子扦样、真实性、净度、发芽率、含水量、生活力及千粒重等7项指标进行研究,确定各指标标准检验方法。结果扦样时种子批的最大总质量可选为5000 g,送检样品的质量最少为150 g;真实性可利用岗梅种子外观结构、大小和颜色等形态特征进行鉴定;净度分析可按照GB/T 3543.1-1995《农作物种子检验规程总则》中的相关方法进行;发芽试验条件为50 mg·L^(-1)赤霉素浸种24 h后于20℃、光照10 h,砂上发芽;种子水分测定采用高恒温[(130±2)℃]烘干法烘2 h;生活力测定采用四氮唑(TTC)染色法,浸种时间为4 h,种子横向切去1/3后于40℃、0.5%TTC溶液中避光染色24 h;质量测定采用百粒法。结论确定了可控指标检验的最优条件,本规程可用于岗梅种子的质量检验。Objective To establish quality- testing methods for Ilex asprella seeds. Methods By referring to the International Seed Testing Rules made by International Seed Testing Association(ISTA) and the Seed Testing for Crops(GB/T3543- 1995) issued by China, methods for seed sampling, genuineness, purity, germination percentage,moisture content,viability and one- thousand- grain weight of Ilex asprella seeds were studied. Results The maximum total mass of seed lot was 5000 g for sampling, and the minimum mass of submitted sample was 150 g. Morphologic identification was used to determine seed genuineness. Purity determination referred to the Seed Testing for Crops(GB/T3543- 1995) issued by China. The germination should be conducted on sand bed at 20 ℃,with 10 h light per day after treatment with 50 mg·L^(-1) gibberellic acid. The seed moisture content was determined by oven drying method at130 ℃ for 2 h. Following 4 h immersing in dd H_2O and 1/3 of transverse cutting,seed viability was tested by soaking seeds in 0.5 % TTC solution for 24 h in dark. One- hundred- grain weight was used to determine the weight of the seed.Conclusion The seed testing rules of Ilex asprella have been established.
