摘  要:目的:观察3'-大豆苷元磺酸钠(DSS)对CCl4所致小鼠肝损伤的保护作用,并研究其对免疫应答的调节作用。方法:以腹腔注射10%CCl_4制作小鼠肝损伤模型。造模期间随机分组,每组分别给予生理盐水、联苯双酯、DSS灌胃治疗,持续6周。检测血清中天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)活性及IL-2、TNF-α含量;流式细胞仪测定全血中T细胞亚群(CD3^+、CD4^+、CD8^+)水平。结果:给予不同剂量DSS治疗肝损伤模型小鼠后,小鼠血清ALT、AST及TNF-α水平均不同程度降低,IL-2水平升高,降低或升高的差异均以高剂量组更为显著(P<0.05或P<0.01);小鼠全血CD3^+、CD4^+细胞比例降低,CD8^+细胞比例提高,CD4^+/CD8^+T细胞比值降低。结论:DSS对CCl_4诱导的慢性肝损伤有保护作用,其作用是通过平衡T淋巴细胞亚群,调节免疫功能实现的。Objective: To explore the protective effects of 3'- daidzein sulfonate sodium( DSS) on chronic liver injury induced by carbon tetrachloride( CCl_4) in mice and to appraise the role of T lymphocyte subsets. Methods: 10% CCl_4 was injected in mice by intraperitoneal injection to make liver injury model. At the same time,the mice were divided into different groups and were treated with normal saline,bifendate and DSS respectively for six weeks,once a day. The activity of AST and ALT,and the level of IL-2 and TNF-α in serum were detected. The T lymphocyte subsets( CD3+、CD4+、CD8+) in blood were observed by using Flow cytometry. Results: Compared with the model group,DSS treated groups showed decrease in the activities of ALT and AST,and the level of TNF-α,and an increase in IL-2 level. After treated with DSS,the ratio of CD3+、CD4~+、CD4+/ CD8+were reduced,while CD8+cell were elevated. Conclusion: DSS plays a protective role in chronic hepatic injury induced by CCl_4 and it is the result of DSS modulating the immunologic function by counterbalance the T lymphocyte subsets.
