葛根素对糖尿病大鼠视网膜中IGF-1和TNF-α表达的影响

摘　　要：目的探讨不同剂量葛根素对糖尿病大鼠视网膜中IGF-1和TNF-α表达的影响。方法将90只健康清洁级Wistar雄性大鼠随机分为5组,分别为对照组(10只)、糖尿病模型组(20只)、葛根素2mg/kg给药组(A组,n=20只)、葛根素5mg/kg给药组(B组,n=20只)和葛根素10mg/kg给药组(C组,n=20只)。除对照组外,其它4组均腹腔注射50mg/kg链脲佐菌素制备糖尿病模型;给药方式均采用每日上午灌胃1次,3个月后处死,取各组大鼠眼球视网膜。采用HE染色检测大鼠视网膜的病理改变;采用免疫组织化学方法检测大鼠视网膜组织中IGF-1、TNF-α的蛋白阳性表达强度。结果与对照组相比较,糖尿病组和A组IGF-1蛋白的阳性表达强度均明显降低、TNF-α蛋白的阳性表达强度均明显升高,差异显著有统计学意义(P<0.01);B组IGF-1蛋白的阳性表达强度降低、TNF-α蛋白的阳性表达强度升高,差异亦有统计学意义(P<0.05);C组IGF-1、TNF-α蛋白的阳性表达强度略降低及略升高,差异无统计学意义(P>0.05)。与糖尿病组比较,A组差异无统计学意义(P>0.05);B组、C组差异均有统计学意义(P<0.01)。结论葛根素能够通过提高IGF-1的表达及降低TNF-α的表达,从而起到保护视网膜的功能。Objective To investigate the effects of Puerarin on the expression of different doses of IGF-1and TNF-αin retina in diabetic rats.Methods 90 healthy clean grade Wistar male rats were randomly divided into 5groups,which were control group（10rats）,diabetic model group（20rats）,puerarin 2 mg/kg gives the medicine group（A group,n=20only）,puerarin 5mg/kg gives the medicine group（B group,n=20only）and puerarin 10mg/kg the drug group（group C,n=20）.Except the control group,diabetic model of inhibin preparation of other 4groups were injected50mg/kg STZ;Administration methods were adopted in the morning,1times in the morning,3months later,the rats were sacrificed.HE staining was used to detect the rat retina pathological change;protein positive expression intensity of IGF-1,TNF-αimmunohistochemical method was used to detect rat retinal tissues.Results Compared with the control group,diabetes group and a group of IGF-1protein positive expression strength were significantly lower,TNF-αprotein expression intensity was significantly higher,the differences of statistical significance（P〈0.01）,reduce the positive expression intensity of IGF-1protein in group B and TNF-αprotein positive expression intensity increased,the difference is also statistically significant（P〈0.05）,C group IGF-1,TNF-αprotein expression intensity decreased slightly and slightly increased,the difference was not statistically significant（P〉0.05）.Compared with diabetic group,agroup without statistical significance（P〉0.05）;B group and C group,the differences were statistically significant（P〈0.01）.Conclusion Puerarin can raise the expression of IGF-1and decrease the expression of TNF-α,so as to protect the function of the retina.
