摘 要:目的探讨清脑止痛胶囊(QNZTJN)对硝酸甘油致偏头痛大鼠的影响。方法健康雄性Wistar大鼠90只,随机分为6组:空白组、模型组、清脑止痛胶囊低(QNZTJN-L,0.35g/kg)、中(QNZTJN-M,0.7g/kg)、高(QNZTJN-H,1.4g/kg)剂量组、阳性药正天丸组(0.006g/kg),每组15只。给药7天后,用皮下注射硝酸甘油(NTG,10mg/kg)复制大鼠偏头痛模型,观察注射硝酸甘油后180min内大鼠行为学反应,造模4h后检测血清及脑干中一氧化氮(NO)、一氧化氮合酶(NOS)、5-羟色胺(5-HT)、降钙素基因相关肽(CGRP)、β-内啡肽(β-EP)含量的变化。结果与对照组比较,模型组大鼠挠头次数明显增加(P<0.05,P<0.01),血浆和脑组织NO含量明显升高(P<0.05,P<0.01),脑组织中NOS、CGRP含量明显升高(P<0.05),β-EP、5-HT含量明显降低(P<0.05);与模型组比较,清脑止痛胶囊中、高剂量组大鼠挠头次数明显减少(P<0.05,P<0.01),血浆和脑组织NO含量明显降低(P<0.05,P<0.01),脑组织中NOS、CGRP含量明显降低(P<0.05),β-EP、5-HT含量明显升高(P<0.05),且呈剂量依赖性。结论清脑止痛胶囊可以缓解大鼠偏头痛症状,其机制可能与调节单胺类神经递质释放,进而改善脑及血管的功能障碍有关。Objective To observe the effect of QNZTJN on rats model of migraine induced by nitroglycerin.Methods Totally 90 Wistar rats(males)were randomly divided into 6groups:Control group,QNZTJN low dose gourp(0.35g/kg),QNZTJN medium dose group(0.75g/kg),QNZTJN high dose group(1.4g/kg),ZTW group(0.006g/kg).The migraine models were induced by injecting 10mg/kg nitroglycerin into the rats to observe the effects of different concentrations of QNZTJN on the level of NO,NOS,5-HT,CGRP,β-EP in the brain tissue and plasma.ResultsCompared with the normal group,the forelimb scratching frequency of rats were increased significantly in model group(P0.05,P0.01),the content of NO in both brain and serum was increased(P0.05,P0.01),the contents of CGRP and NOS in brain were increased(P0.05),but the contents ofβ-EP and 5-HT in the brain were decreased significantly(P0.05,P0.01).Compared with model group,QNZTJN medium and high dose groups significantly reduced the forelimb scratching frequency of rats(P0.05,P0.01),the content of NO in both brain stem and serum was decreased(P0.05,P0.01),the contents of CGRP and NOS were decreased(P0.05),but the contents ofβ-EP and 5-HT in the brain stem were increased(P0.05,P0.01).These effects were in a dose dependent manner.Conclusion The QNZTJN can relieve migraine attacks,which may regulate the release of monoamine neurotransmitters,thus improving the function of the brain and vascular disorders.
