摘 要:目的研究姜黄素(curcumin,CUR)对肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)刺激的INS-1细胞功能损害的影响及可能的作用机制。方法培养INS-1大鼠胰岛瘤细胞,不同浓度(0、0.3125、0.625、1.25、2.5、5、10、20、40、80、160μmol/L)CUR作用24 h后,用MTT方法检测生长状态,用显微镜观察细胞形态。Real-time PCR法测定CUR对TNF-α刺激的INS-1细胞胰岛素m RNA表达的影响。ELISA法测定CUR对TNF-α刺激的INS-1细胞胰岛素分泌的影响。Western Blot法检测CUR对TNF-α刺激的INS-1细胞脂联素受体1(adiponectin receptor-1,Adipo R1)和葡萄糖转运蛋白2(glucose transporter-2,Glut2)蛋白的表达。结果 20μmol/L以下浓度的CUR对INS-1细胞无毒性作用,对INS-1细胞形态无影响。CUR改善了TNF-α刺激的INS-1细胞胰岛素m RNA表达和胰岛素分泌减少,在CUR浓度为10μmol/L时作用效果最为显著(P<0.05)。CUR改善了TNF-α刺激的INS-1细胞Adipo R1、Glut2蛋白表达降低(P<0.05)。结论 CUR可改善TNF-α对INS-1细胞胰岛素合成和分泌的抑制作用,其作用机制可能与脂联素受体通路有关。Objective To investigate the effect of curcumin(CUR) on pancreatic beta–cell line INS-1 stimulated by tumor necrosis factor-α(TNF-α) and the possible mechanism. Methods INS-1 cells were cultured with different concentrations of CUR(0,0.3125,0.625, 1.25,2.55,10,20,40,80,160 μmol/L) for 24 h. The cell viability was determined by MTT assay and the cell morphological changes were observed under microscope. The expression of insulin m RNA and insulin secretion were measured by real-time PCR and ELISA respectively, and the protein expression of adiponectin receptor-1(Adipo R1) and glucose transporter-2(Glut2) were measured by Western blot. Results CUR at the concentration lower than 20 μmol/L had no toxicity on INS-1, nor did it change the cell morphology. In addition, CUR improved the decreased expression of insulin m RNA and insulin secretion in INS-1 cells treated by TNF-α, especially at CUR concentration of 10 μmol/L(P〈0.05). Moreover, CUR improved the protein expression of Adipo R1 and Glut2(P〈0.05). Conclusion CUR could improve the decreased insulin secretion and insulin m RNA expression in INS-1 cells induced by TNF-α, and the effects may be related to the adiponectin receptor pathway.
