Apelin-13对血清剥夺诱导的人脐带间充质干细胞凋亡的影响

摘　　要：目的观察apelin-13对无血清培养诱导的人脐带间充质干细胞(h UC-MSCs)凋亡的影响。方法采用组织块贴壁培养法培养h UC-MSCs,随机分成无血清培养组(A组)和不同浓度apelin-13处理组,apelin-13处理组浓度分别为0.1 mmol/L(B1组)、0.5 mmol/L(B2组)、5 mmol/L(B3组)、10 mmol/L(B4组)。倒置显微镜观察细胞形态;流式细胞仪测定h UC-MSCs表面免疫标记;流式细胞术检测各组细胞凋亡率,MTT法检测0.5、5 mmol/L apelin-13对h UC-MSCs增殖活性影响。结果流式细胞仪鉴定结果表明,h UC-MSCs表达CD44、CD90、CD105、CD73、HLA-ABC,不表达CD34、CD45、HLA-DR。与A组比较,B1、B2、B3、B4组均抑制无血清培养诱导的h UW-MSCs早期凋亡,凋亡率比较差异均有统计学意义(P<0.05),B3、B4组与B1、B2组间差异有统计学意义(P<0.05),B3组与B4组、B1组与B2组差异无统计学意义(P>0.05)。A组与B2、B3组比较,细胞在490 nm OD值差异无统计学意义(P>0.05)。结论一定浓度内的apelin-13能抑制无血清培养诱导的h UC-MSCs早期凋亡,且这种抗凋亡作用可能不是通过促增殖作用引起的。Objective To investigate the effect of apelin - 13 on apoptosis of human umbilical cord mesenchymal stem cells（hUC -MSCs） induced by serum -free culture. Methods hUC -MSCs were cultured by tissue adherent culti- vation method and randomly divided into four groups ： the serum - free culture group （ Group A） and different concentrations of apelin - 13 treatment groups （ B1, B2, B3, B4, treated with apelin - 13 at 0. 1, 0. 5, 5, 10 mmol/L, respective- ly）. The morphological changes were observed by the inverted microscope. Immunophenotypic analysis of cell surface markers was performed by flow cytometry. Cell apoptosis rate was also examined by flow cytometry. The effects of O. 5, 5 nM apelin - 13 on hUC - MSC proliferation were determined by MTT method. Results hUC - MSCs expressed CD44, CDg0, CD105, CD73 and HLA - ABC, while did not express CD34, CIM5 or HLA - DR. Compared with Group A, early apoptosis of hUW - MSCs was inhibited in Groups B1, B2, B3 and IM （ P 〈 0.05 ）. The differences between B3, 1t4 and B1, B2 were statistically significant. In the 490 -nm OD value, Group A and Groups B2, B3 were similar （P 〉 0. 05）. Conclusion In a certain range of concentration, the apelin - 13 could inhibit the early apoptosis of hUC - MSCs cultured by serum - free medium, and this anti - apoptosis effect may not be caused by promoting proliferation.
