摘  要:以阳丰甜柿(Diospyros kaki cv.Youhou)的带腋芽茎段为外植体,研究了植物生长调节剂及其浓度配比对其腋芽的诱导、增殖及生根的影响。结果表明,带芽茎段接种在1/2MS+1.0mg·L^(-1)ZT+0.1mg·L^(-1)IAA培养基培养4周后,腋芽诱导率为77.8%。将萌发的腋芽切成带芽茎段或单株转到1/2MS+1.0mg·L^(-1)ZT+2.0mg·L^(-1)2ip+0.1mg·L^(-1)IAA培养基继代培养,最高增殖系数可达4.1。在增殖培养中长至3cm以上的芽转入1/2MS+0.75mg·L^(-1)IBA的生根培养基中暗培养7d后,转入光下培养30d,生根率可达73.3%,平均生根数为3.6。该研究建立了阳丰甜柿离体再生体系,为其规模化生产提供了技术平台。Taking the stem segments with auxiliary buds of Diospyros kaki cv.Youhou as explants,influences of plant regulators and their concentrations on the induction,proliferation and rooting were examined.The induction rate of auxiliary bud was 77.8% after the explants were cultured 4weeks in the medium of 1/2MS+1.0Mg·L^(-1)Zeatin+0.1mg·L^(-1)IAA.The regenerated buds were then subcultured in the medium of 1/2MS+1.0mg·L^(-1)ZT+2.0mg·L-12ip+0.1mg·L^(-1)IAA,the highest proliferation coefficient was 4.1.The shoots with 3cm in length were transferred into the medium 1/2MS+0.75mg·L^(-1)IBA,and cultured for 7din dark and 30 din light,the rooting rate was 73.3%,and the average number of root was 3.6.This results established the in vitro propagation system for Diospyros kaki,and provided the technical basis for scale production.
