摘  要:试验以H99、综31和郑58三个玉米自交系的成熟胚为外植体,利用正交试验设计L9(34),对影响愈伤组织诱导的基因型、培养基、2,4-D浓度和Ag NO3浓度4个因素进行了研究。结果表明:各因素对初级愈伤诱导率影响大小为基因型>培养基>2,4-D浓度>Ag NO3浓度;对胚性愈伤诱导率影响大小为基因型>Ag NO3浓度>2,4-D浓度>培养基。诱导中最优组合为综31+10 mg/L Ag NO_3+MN+2 mg/L 2,4-D,继代中最优组合为综31+10 mg/L Ag NO3+MN+1 mg/L 2,4-D,其初级愈伤诱导率为87.41%,胚性愈伤诱导率为29.74%。The maize inbred lines of mature embryos of H99, Zong 31 and Zheng 58 were taken as explants. Four factors which affect callus inducement, including genotype, substrate, 2,4-D concentration and AgNO3, were studied by using orthogonal experiment L9(34). The results showed that the rank of primary callus induction rate was genotype〉 substrate〉 2,4-D concentration 〉AgNO3 concentration and the rank of embryonic callus induction rate was genotype 〉AgNO3 concentration 〉 2,4-D concentration〉 substrate. The best combination in induction was Zong 31+10 mg/L AgNO3 + MN+2 mg/L 2,4-D and the optimal combination in subculture was Zong 31 + 10 mg/L AgNO3 +MN+ 1 mg/L 2,4-D,whose primary callus inductivity rate was 87.41% and embryonic callus inductivity rate was 29.74%.
