摘  要:以21日龄雌性小鼠为研究对象,用雌激素处理一周后收集子宫,苏木素-伊红(HE)染色法观察到子宫发生水肿、内膜外膜明显增厚。提取子宫总RNA和总蛋白,通过实时荧光定量和Western-Blot方法检测雌激素对子宫中神经营养因子BDNF及NGF的表达。荧光定量结果表明:试验组BDNF表达显著升高、NGF表达显著降低。Western-Blot结果与荧光定量结果一致。这说明雌激素能够调节小鼠子宫中BDNF和NGF的表达。Studies showed that neurotrophic factors and estrogen play an important role in regulating animal reproduction. This study took 21-day-old female mice as research object,treating them for a week with estrogen intraperitoneally,and the collected uteruses were stained by Hematoxylin-Eosin( HE). By observation,the study showed that uterine edema was induced,and endometrium and outer membrane of the uteruses were significantly thickened. Total RNA and total protein were extracted for quantitative analysis to explore the effects of estrogen on uteru expression of neurotrophic factors BDNF and NGF by RT-PCR and Western-Blot. The results of RT-PCR indicated that the expression of BDNF in uterus of mice treated with estrogen had a significant increase,while the expression of NGF in uterus of mice treated with estrogen had a significant decrease. The results of Western-Blot showed an accordant trend with that of RT-PCR,suggesting that estrogen may regulate the expression of BDNF and NGF in mice uterus.
