强直性脊柱炎患者外周血单个核细胞踝蛋白1表达的研究

摘  要:目的探讨强直性脊柱炎(AS)患者、类风湿关节炎(RA)患者及健康志愿者(HV)外周血单个核细胞(PBMC)中踝蛋白1的表达水平及其差异。方法采集AS、RA患者及HV外周血样本各30例,流式细胞术分析CD3^+T细胞及CD14^+单核细胞中踝蛋白1蛋白平均荧光强度(MFI),逆转录PCR(RT-PCR)检测PBMC中踝蛋白1 mRNA表达水平,比较组间踝蛋白1在蛋白及mRNA表达的差异。结果在CD3^+T细胞及CD14^+单核细胞中,AS患者的踝蛋白1 MFI水平均高于HV和RA患者(P均<0.05),而HV与RA患者比较差异无统计学意义(P>0.05)。RTPCR提示AS患者、HV及RA患者踝蛋白1 mRNA相对表达水平分别为1.37±0.22、0.26±0.05、0.27±0.06,AS患者的踝蛋白1 mRNA相对表达水平均高于HV和RA患者(P均<0.05)。结论AS患者PBMC中踝蛋白1表达水平明显高于HV及RA患者,提示踝蛋白1可能在AS发病机制中发挥作用。Objective To investigate the expression of Talin 1( TLN 1) in peripheral blood mononuclear cell( PBMC) among patients with ankylosing spondylitis( AS),rheumatoid arthritis( RA) and healthy volunteer( HV). Methods Collect samples of peripheral blood in 30 patients with AS,HV and RA respectively. The mean fluorescence intensity( MFI) of TLN 1 in the CD3^+T lymphocytes and CD14^+monocytes were analyzed by flow cytometry. Also,the mRNA expression of TLN 1 in PBMC was detected by the reverse transcription- polymerase chain reaction( RT-PCR). The protein and mRNA level of TLN 1 were compared among AS,RA and HV groups. Results MFI of TLN 1 in CD3^+T lymphocytes and CD1+monocytes of AS patients were both significantly higher than those of HV and RA patients( P < 0. 05),while there was no statistically significant difference between HV and RA patients( P > 0. 05). Moreover,the relative mRNA level of TLN 1 in patients with AS was 1. 37 ± 0. 22,significantly higher than that of HV and RA patients( 0. 05 ±0. 27 and 0. 26 ± 0. 06 respectively,both P < 0. 05). Conclusion TLN 1 was up-regulated in the PBMC of AS patients,indicated that TLN 1 might play an important role in the pathogenesis of AS.
