摘  要:目的探讨lncRNA TUG1(taurine up-regulated 1)与冠心病的关系,研究TUG1对脐静脉内皮细胞(HUVECs)功能的影响。方法采用real-time PCR检测冠心病病例对照外周血单个核细胞(PBMCs)中TUG1的表达。利用siRNA敲低HUVECs内TUG1表达水平,并检测对细胞凋亡及IL-8表达水平的影响。建立并应用低氧和TNF-α刺激的内皮细胞损伤模型,探讨TUG1在内皮细胞功能紊乱中的作用。结果冠心病患者PBMCs样本中TUG1表达显著高于对照组(P<0.001)。敲低TUG1显著抑制正常培养及低氧或TNF-α刺激诱导的内皮细胞凋亡(P<0.05)。敲低TUG1并予以低氧或TNF-α刺激,IL-8的表达明显下降(P<0.001)。结论 LncRNA TUG1在冠心病患者中出现异常高表达。体外实验结果显示,敲低TUG1能抑制内皮细胞凋亡,降低IL-8的表达水平。提示TUG1可能影响血管内皮细胞功能,参与冠心病的发生发展过程。Objective To explore the potential relationship of IncRNA TUG I(taurine up-regulated 1) in coronary artery disease(CAD) and its biological function in human umbilical vein endothelial cells(HUVECs).Methods The relative expression of TUG1 in peripheral blood mononuclear cells(PBMCs) was detected by real-time PCR in CAD patients and control individuals,respectively.TUG1 was knocked down by specific siRNAs,and the apoptosis of endothelial cells was analyzed by Annexin V-FITC apoptosis detection kit.Results Compared with health control,the expression of TUG1 was significantly increased in PBMCs of CAD patients(P 〈0.001).Down regulation of TUG1 inhibited endothelial cells apoptosis and the apoptosis induced by hypoxia and tumor necrosis factor-α(TNF-α)(P 〈0.05).And the knocking-down of TUG1 reduced the expression of interleukin-8(IL-8) in vitro(P 〈0.001).Conclusion The expression of TUG1 abnormally increased in PBMCs of CAD patients.Furthermore,in vitro assay also demonstrated that the knocking-down of TUG1 inhibited cell apoptosis and reduced the expression of IL-8 in HUVECs.Thus,TUG1 might be involved in endothelial dysfunction and the pathogenesis of CAD.
