芋组培脱病毒技术及其增产效益的研究

摘　　要：试验研究了莱阳孤芋茎尖培养脱毒技术及脱毒芋的产量表现。结果表明 ,茎尖大小在0 3～ 0 5mm时 ,成活率在 80 %以上 ,适宜茎尖分化的培养基为MS +BA 1 0mg/L +NAA 0 2mg/L +Spm 2 0mg/L。 3次剥离茎尖培养所得试管苗经酶联免疫检测及电镜观察显示 ,已不带芋花叶病毒。脱毒试管苗定植后 ,植株生长旺盛 ,后代球茎数量达每株 31 2个。小区对比试验表明 ,脱毒原原种芋、原种芋可使球茎数量比对照增加 37 0 %～ 5 2 5 % ,同时使单位面积产量和商品芋产量分别增加 2 0 2 %～ 4 3 9%和 4 9 9%～ 70 0 %。The virus-eliminating technology by shoot-tip culture was studied on taro (Colocasia esculenta) The results showed that the survival rate of shoot-tip,at 0 3～0 5 mm in diameter,was over 80%; the best medium for shoot differentiation was MS+BA 1 0 mg/L+NAA 0 2 mg/L+Spm 20 mg/L The two methods (the SPA-ELISA and the electron microscope) were used on the virus detection of the plantlets The results indicated that the plantlets did not carry dasheen mosaic virus with exfoliating shoot-tip ( shorter than 0 5 mm) culture by three times The seedlings were growing luxuriantly after virus-free plantlets were planted in the net-room,and the cormel amount was upto 31 2 per plant The results of the plot test showed that,as compared with the control,the amount of cormels of virus-free taro had increased by 37 0%～52 5%,the yield had increased by 20 2%～43 9%; and the weight of the commodity cormels (more than 30 g per cormel) had increased by 49 9%～70 0%
