摘 要:探索了镍离子对人血管内皮细胞增殖的影响。本研究以人脐静脉内皮细胞为研究对象,从细胞生长活性、细胞形态等几个方面入手,运用MTT、姬姆萨染色等方法探索不同浓度的镍离子对血管内皮细胞增殖的影响。通过研究发现,氯化镍对人脐静脉内皮细胞的半数致死浓度为260 mg/L。在镍离子作用下,HUVEC细胞增殖受到抑制,细胞形态有中毒样改变,在观察浓度及时间范围内,呈现浓度依赖和时间依赖趋势。The influence of nickel ions on the proliferation of HUVEC cells( Human Umbilical Vein Endothelial Cells) was evaluated. MTT and Giemsa stein were used to assess the cell viability and cell morphology under different concentrations of nickel ions. According to our research,the lethal dose 50 of Nickel Chloride was 260 mg / L. Inhibition rate increased with the increasing of concentrations of nickel ions and also increased with the time exposure to nickel ions.Ultrastructural alterations were observed including irregular shaped nuclei and the phenomena were dose dependent.Cellular proliferation of HUVEC was obviously inhibited by nickel ions. Cellular morphology of HUVEC became to be toxic state,and that was dose dependent and time dependent.
